摘要 目的:研究驱动蛋白Rbkinesin-6对人肺腺癌细胞系A549有丝分裂的影响。方法:采用Rbkinesin-6特异性esiRNA介导的RNAi技术,系统分析Rbkinesin-6对肺癌细胞系A549有丝分裂的影响。结果:Rbkinesin-6esiRNA可特异性地消除A549细胞Rbkinesin-6的表达,Rbkinesin-6缺失的细胞无法完成有丝分裂末期/胞质分裂,进而转变为双/多核细胞。结论:Rbkinesin-6在A549细胞有丝分裂末期/胞质分裂最后阶段起重要作用,Rbkinesin-6基因沉默可以抑制肺癌细胞的生长,为肺癌基因治疗的研究提供了新的思路。 Objective: To study the function of Rbkinesin-6 in mitosis in human pulmonary adenocarcinoma cell line A549. Method: esiRNA technique was used to systematically analyze the function of Rbkinesin-6 protein in mitosis in A549 cells. Results: Rbkinesin-6 esiRNA could effectively and specifically ablate the expression of Rbkinesin-6. Rbkinesin-6 ablated cells could not finish the telophase/cytokinesis phase, and became binuclear/multinuclear cells. Conclusion: Rbkinesin-6 plays an important role in telophase/cytokinesis phase, Rbkinesin-6 ablation can inhibit the growth of lung cancer cells.
