摘要
目的通过抑制肝癌细胞中KIF14表达,观察紫杉醇对肝癌细胞增殖的影响。方法应用RNA干扰技术,肝癌细胞中沉默KIF14表达,在经紫杉醇处理细胞后采用MTT试验检测肝癌细胞增殖能力的变化。结果肝癌细胞中沉默KIF14的表达可以增强其对紫杉醇的敏感性。结论 KIF14在肝癌细胞耐药发生过程中发挥着重要作用,进一步研究其作用机制可以为寻找克服肝癌耐药的有效途径提供理论依据。
Objective This study observes the effect of paclitaxel on liver cancer cell proliferation by inhibiting KIF14 expression in liver cancer cells. Methods Silenced KIF14 expression in liver cancer cells by RNA interference,these cells were delt by paclitaxel and the proliferation ability of liver cancer cells was determined by MTT test. Results Silence KIF14 expression in liver cancer cells enhanced the cells sensitivity to paclitaxel. Conclusion KIF14 is important in the process of drug resistance produced by human hepatocellular carcinoma cells. Further study on its mechanism can provide theoretical basis for seeking the effective ways to overcome drug resistance of liver cancer.
