摘要
为了提高血吸虫病病因诊断的特异性和敏感性,用慢性日本血吸虫病病人血清筛选以λgtll构建的日本血吸虫中国大陆株成虫cDNA表达库,获得一阳性克隆Sjgt4,其表达产物为148000融合蛋白。插入的cDNA片段经聚合酶链反应(PCR)法扩增、纯化后,亚克隆入高表达载体pGEX,获得克隆SjpG5,其表达产物为58000融合蛋白,应用免疫印迹技术,其可被96%(24/25)的日本血吸虫病病人血清中特异抗体所识别。
he sera of patients with chronic infection of
Schis-tosoma japonicum (Sj) were used for screening thecDNA expression library constructed
from Sj adultworm, and an antigen-positive clone Sjgt4 which ex-pressed an 148 KD fusion
protein was identified. Theinserted Sj cDNA fragment was amplified by PCR andsubcloned into
the vector PGEX with high level of ex-pression. A positive clone SjpG5 which expressed a
fu-sion protein with a molecular weight of 58 KD was ob-tained. When the fusion protein was
used for detectionof Sj patients’ sera by Western blpt, 24 out of 25 serawere positive , a
positive rate of 96 %.
