摘要
谷物样品不进行预处理而采用Douglas法直接测定其中的戊聚糖含量在实际应用中存在很大的误差,文中比较不同预处理方法对多种原料中戊聚糖含量测定结果的影响,确定不同原料的最佳预处理方法,对复杂体系水解后可能产生的单糖对测定结果的影响也进行了初步的研究。结果表明:在实际情况中,应用Douglas方法前必须对不同原料进行不同的预处理,复杂体系经水解后可能产生的单糖中六碳糖对木糖的最大吸收峰几乎没有影响,对谷物样品进行完全降解预处理,所得检测结果与实际结果相差较小。另外,研究发现对固体样品采用硫酸法和酶法进行预处理得到的结果准确度相当,而对于液体样品酶法预处理明显优于其他方法。
Measuring of pentosans is very important when detecting cereal samples. The typical pentosans determination method 'Douglas' was applied directly without any special pretreatment of the samples, thus causing a big different between the real content of pentosans in the raw material and the test result from Douglas method. Dextose which might produced during hydrolyzing the complex material had little influence on the results. Different pretreatments of the samples were compared, and the best feasible preparation for the different kinds of samples were obtained. The results displayed that different methods are needed before apply of Douglas method. For solid samples, both vitriol method and the enzyme method were work fine, while in the liquid samples, enzyme method was better.
