The protective effect of curcumin on energy metabolism in rat myocardial cells against ischemia and reperfusion

摘要 目的:探讨姜黄素对大鼠缺血再灌注损伤心肌能量代谢的影响。方法:建立Langendorff灌注模型,将32只Wistar大鼠随机分为对照组和姜黄素组。对照组心脏停搏液用4℃StThomas液;姜黄素组在4℃StThomas液中加入姜黄素(0.2mmol·L)。分别于缺血前及再灌注30min后,检测心脏血流动力学指标、心肌线粒体超氧化物歧化酶(SOD)、丙二醛(MDA)含量。31P核磁共振分光仪检测心肌磷酸肌酸(PCr)、磷酸盐(Pi)和β-ATP水平。透射电镜观察再灌注后线粒体超微结构。结果:姜黄素组心肌再灌注30min后心脏血流动力学指标、心肌高能磷酸盐水平、线粒体MDA、SOD、线粒体超微结构等均明显优于对照组(P<0.01)。结论:姜黄素可显著改善大鼠缺血-再灌注损伤心肌的能量代谢,与其增强心肌线粒体内源性抗氧化能力,维持线粒体结构的完整性有关。 OBJECTIVETo study the effect of curcumin on energy metabolism in rat myocardial cells against ischemia and reperfusion.METHODSThirty-two rats were randomly divided into two groups. Curcumin group(n=16),isolated rat hearts were preserved in (4 ℃) St Thomas solution with curcumin ((0.2) mmol·L^(-1)) for 2 hours. Control group(n=16), isolated rat hearts were preserved in (4 ℃) St Thomas solution for 2 hours. Then the hearts were mounted on Langendorff model. Before and after 30 min of reperfusion, the hemodynamic parameters, the mitochondrial superoxide dismutase (SOD), malodiadehyde (MDA) were investigated. Transmission electron microscopy was used to examine the microstructural changes of mitochondrial after 30 min of reperfusion. Phosphocreatine (PCr), inorganic phosphate (Pi) and β-adenosine triphosphate (β-ATP)were measured using ()^(31)p-nuclear magnetic resonance (NMR) spectroscopy. RESULTSRecovery of cardial function and mitochondrial ultrastructure in Curcumin group was significantly better than those in control group. Mitochondrial malodiadehyde (MDA) in Curcumin group were significantly lower than that in control group (P<(0.01)). Mitochondrial superoxide dismutase (SOD) was significantly higher than that in control group (P<(0.01)),as well as the myocardial PCr, Pi and β-ATP content. CONCLUSIONCurcumin could significantly improve the energy metabolism of myocardial cells against ischemia and reperfusion. Its protective effect maybe correlate with the decrease of oxgen free radical in the mitochondrial and the preservation of high energy phosphate as well as the maintance of integration of myocardial mitochondrial ultrastructure.
