摘要
目的 观察中药姜黄的主要活性单体成分双脱甲氧基姜黄素(姜黄素Ⅲ)和顺铂联用对抑制人肺腺癌细胞系A5 49裸小鼠移植瘤生长有无增效作用。方法 将2 0只荷人A5 49肺腺癌的裸小鼠随机分成4组:①阴性对照组;②姜黄素Ⅲ组(10 0mg kg ,隔日1次×8次) ;③顺铂组(5mg kg ,每周2次×4次) ;④联合用药组(姜黄素Ⅲ10 0mg kg ,隔日1次×8次+顺铂5mg kg ,每周2次×4次)分别测量瘤质量、瘤体积和微血管密度,并观察各组移植瘤组织的病理形态结构。结果 4个组瘤质量分别为(1 15±0 18)、(0 5 7±0 10 )、(0 49±0 0 8)、(0 2 4±0 0 5 )g ;瘤体积分别为(14 13 4±2 66 3 3 )、(699 40±118 0 3 )、(5 90 88±114 0 5 )、(3 5 4 72±12 3 72 )mm3。统计学分析表明:在瘤质量及瘤体积方面,姜黄素Ⅲ组、顺铂组和阴性对照组比较差异具有显著性(P <0 0 1) ,联合用药组和其他3组比较均有显著性差异(P <0 0 1)。结论 姜黄素Ⅲ、顺铂对A5 49裸小鼠移植瘤生长都有明显的抑制作用;姜黄素Ⅲ和顺铂联用能起增效作用。
Objective To study whether curcumin Ⅲ could potentiate the anti-tumor effect of cisplatin on xenografted tumor growth of human lung adenocarcinoma cell line A549 in nude mice. Methods Twenty nude mice bearing human lung adenocarcinoma A549 xenografted tumor were randomly divided into 4 groups (n=5 in each group): control group (0.2 ml mixture of 6% dehydrated alcohol and 6% macrogol 400, intraperitoneal injection once every other day, 8 times), curcumin Ⅲ group (100 mg/kg, intraperitoneal injection once every other day, 8 times), cisplatin group (5 mg/kg, intraperitoneal injection twice a week, 4 times), the combination group of curcumin Ⅲ and cisplatin (the dose of curcumin Ⅲ and cisplatin was the same with that of curcumin Ⅲ group and cisplatin group, respectively). Results The mean tumor weights of 4 groups were (1.15±0.18), (0.57±0.10), (0.49±0.08), (0.24±0.05) g, respectively and the mean tumor volumes (1 413.4±266.33), (699.40±118.03), (590.88±114.05), and (354.72±123.72) mm 3, respectively. The difference in the tumor weight and volume between the control and curcumin Ⅲ group as well as the control and cisplatin group were both significant (P<0.01). The tumor weight and volume of the combination group were significantly different from those of other 3 groups (P<0.01). Conclusion Curcumin Ⅲ and cisplatin can significantly inhibit tumor growth in nude mice bearing human lung adenocarcinoma cell line A549 and their combination can produce a synergistic action.
