摘要
目的:帕金森病是临床上最常见的神经退行性疾病之一,其确切的病因和发病机制目前尚不清楚。目前一般认为该病主要是由于环境和/或遗传所导致的线粒体功能受损及α-突触核蛋白异常聚集所致。线粒体功能受损尤其表现在complexⅠ功能下降,线粒体DNA的变化,氧应激和凋亡的发生,文章将重点阐述以上内容的研究新进展,以期寻找线粒体损伤和α-突触核蛋白在帕金森病中的确切作用。资料来源:应用计算机检索Medline数据库1980-01/2004-06期间的相关文章,检索词为“pakinson'sdisease和“α-synuclein,mitochondria,complexⅠ,oxidativestress,mtDNA,apoptosis,分别地组合进行检索,限定文章语言种类为英文。同时计算机检索中国期刊全文数据库1999-01/2004-06期间的相关文章,检索词“α-突触核蛋白,线粒体,复合酶Ⅰ,氧应激,线粒体DNA,凋亡,限定文章语言种类为中文。资料选择:对资料进行初审,从查到的资料中选取与α-突触核蛋白,线粒体在帕金森病发病机制中的作用有关的文章。资料提炼:共收集到62篇符合上述要求的文章,查找全文,有48篇文章找到了全文。将观点非常相似的文章进行对比,筛出阐述详尽,近期发表的文章共26篇,其余排除。资料综合:将所选文章按其所述机制与α-突触核蛋白,复合酶Ⅰ,氧应激,线粒体DNA,凋亡的?
OBJECTIVE:Parkinson disease(PD) is one of the most frequent neurodegenerative disorders.The exact aetiology and pathogenesis of PD is still unknown.In recent years,it has become increasingly clear that PD involves in dysfunction of mitochondria and aggregation of α - synuclein induced by environment or inheritance.The dysfunction of mitochondria includes complexⅠ deficiency,mutation of mtDNA,oxidative stress and apoptosis.To introduce the recent development of above contents in this review.In order to find the role of dysfunction of mitochondria and α - synuclein in PD.DATA SOURCES:Using the key terms “ Parkinson disease' combined with “ α - synuclein,mitochondria,complexⅠ ,oxidative stress,mtDNA,apoptosis” respectively, we searched the MEDLINE database for the relevant articles published from January 1980 to June 2004 in English.Meanwhile,using the key terms “ α - synuclein,mitochondria,complexⅠ ,oxidative stress,mtDNA, apoptosis” ,we searched the Chinese journals full- text database from January 1999 to June 2004 in Chinese.STUDY SELECTION:All articles were selected firstly,and those associated with the roles of α - synuclein and mitochondria in pathogenesis of PD were selected.DATA EXTRACTION:A total of sixty- two articles according with above requirements were collected and the full text was looked up.Forty- eight among them were found corresponding full texts.The articles were read and those had the similar views were compared.Ttwenty- six recent published and reasoned articles were collected and the rest were excluded.DATA SYNTHESIS:According to different relativities between different mechanisms described in above articles and α - synuclein,complex I,oxidative stress, mitochondrial DNA,apoptosis,these articles were classified. CONCLUSION:Dysfunction of mitochondria and α - synuclein make important roles in Parkinson disease.Dysfunction of mitochondria leads to an inhibition of ATP synthesis and deficient energy, aggregation of α - synuclein, and cell death.
