摘要
目的:观察高糖对体外培养的大鼠施万细胞半乳糖神经酰胺转移酶(CGT)和半乳糖神经酰胺(GalC)表达的影响。方法:体外培养大鼠施万细胞,用MTT法比较正常葡萄糖(30 mmol·L-1)和高糖(50 mmol·L-1)培养24 h施万细胞的存活力,用半定量RT-PCR 方法测定细胞CGT mDNA表达,用免疫细胞化学方法测定细胞GalC的表达。结果:高糖培养24 h,施万细胞存活力较正常葡萄糖培养时明显下降(73.9%vs 100%,P<0.001);高葡萄糖培养的施万细胞CGT mRNA表达上调:GalC阳性颗粒较正常葡萄糖培养时明显增多。结论:高葡萄糖时施万细胞CGT和GalC表达上调,可能影响施万细胞的活力。
Aim: To observe the expressions of UDP-galactose: ceramide galactosyltransferase ( CGT ) and galactocerebrosides(GalC) in Schwann cells cultured in high glucose. Methods: Schwann cells derived from postnatal rats were cultured in vitro. Survival of Schwann cells both in normal glucose(30 mol·L-1)and in high glucose(50 mol·L-1) was assessed by MTT CGTmRNA expressions in Schwann cells was measured by RT-PCR,and GalC expressions was observed by immunocytochemistry. Results: Survival of Schwann cells in high glucose was decreased significantly when compared with that in normal glucose (73.95% vs 100%), P < 0.01. CGTmRNA expression of Schwann cells in high glucose was raised significantly. GalC expressions of Schwann cells in high glucose was also increased significantly. Conclusion: Both CGT expression and GalC expression were increased significantly in Schwann cells cultured in high glucose.
