摘要
用抗Ⅳ型胶原单克隆抗体免疫组化法对实验性铬酸钠中毒大鼠肾进行了研究。结果显示：对照组肾组织各部基膜Ⅳ型胶原免疫组化染色呈弱阳性。一次气管内注入铬酸钠０．２ｍｇ／ｋｇ２天后，在肾组织出现损伤的同时，其染色亦增强呈阳性。一次气管内注入铬酸钠０．９８ｍｇ／ｋｇ后第２至２８天其染色均较对照组增强，以第４至２１天为甚。Ⅳ型胶原在肾组织各部基膜的染色强弱与肾组织损伤程度相关。结果提示Ⅳ型胶原可作为一个指标来评价铬等其它重金属所致肾损伤的发生、发展和转归。
The rat kidneys of sodium chromate intoxication were studied by immunohistochemical method using the monoclonal antibody to type Ⅳ collagen. The weak positive immunostaining by anti-type Ⅳ collagen antibody was observed along the basement membrane of different sites in normal kidney. The kidney damage and the positive immunostaining were observed in the kidneys of experimental group on the third day after intratrachea injection at dose of 0.2mg/kg sodium chromate. The positive and the strong positive immunostaining were observed in the kidneys of experimental groups on days 4 , 7, 14 , 21 and 28, respectively ,after intratrachea injection at dose of 0. 98mg/kg sodium chromate. The stronger the immunostaining was , the more serious the kidney damage was.The results suggested that type Ⅳ collagen could be used a marker to assess the occurence, development and prognosis of the kidney damage caused by chromium or other heavy metals.
