介绍放射性同位素标记核酸的方法,包括缺口平移法标记DNA、引物延伸法标记DNA、聚合酶法标记RNA、T4多核苷酸激酶标记DNA或RNA的5′-末端、末端脱氧核苷酸基转移酶标记DNA的3′-末端、DNA聚合酶ⅠKlenow片段标记DNA的3′-末端、T4DNA聚合酶标记DNA的3′-末端、标记寡核苷酸。
Nucleic acid probe labelled with radioisotope is a useful tool in research of gene engineering and molecular biology. In this review, the methods for incorporating radioisotopes into nucleic acid were described. They include the nick-translation, primer extension, RNA polymerase-based methods, end-labelling ( 5′-end-labelling with T4 polynucleotide kinase, 3′-end-labelling with terminal deoxynucleotidyl transferase, end repair catalysed by Klenow polymerase, labelling with T4 DNA polymerase) , labelling of oligonucleotides.
