摘要
在抗DNV的品种秋丰、高感品种华八 35及其近等基因系BC6 中,通过 495个RAPD随机引物在各个品种的DNA混合物中进行PCR扩增,获得了一个与家蚕抗DNV基因相关的分子标记S366,对这个标记进行了克隆、测序,并且根据序列设计特异引物转换成SCAR标记,在多个敏感性和抗性品种中进行了验证,证明此分子标记真实可靠。通过生物信息学对测序片段进行分析,发现该片段序列与黄嘌呤脱氢酶基因有 91%的同源性。
In our study, two silkworm strains, namely Qiufeng which is resistant to DNV and Huaba35 which is highly susceptible to DNV and their isogenic line BC_6, were used for screening molecular markers by using 495 RAPD random primers in DNA mixtures of these strains respectively.One molecular marker named S366 was found to link with major gene resistant to DNV,then it was cloned and sequenced.According the sequence, special primer was synthesized and tested in several silkworm strains which are highly susceptible or resistant to DNV.Based on bioinformatic analysis, we found S366 has 91% homology to xanthine dehydrogenase gene.
