摘要
对灯盏花遗传转化中的组织培养受体系统进行试验研究 ,发现以MS为基本培养基 ,使用BA0 2 +NAA2 0的激素配比及浓度 ,播种期 10d的幼苗 ,取真叶作为外植体 ,大小 0 5× 1 5cm2 ,能在二周内较快形成愈伤组织。使用MS +BA0 5+NAA0 5培养基、加 3 0 g/L的蔗糖 ,以真叶为外植体 ,能在二周内较快诱导不定芽形成。使用 75 μg/ml的chl作为土壤农杆菌转化培养的选择压力 ,使用 5 0 μg/ml的Gent作为转基因植株的选择压力 ,使用 3 0 0 μg/ml的Cef作为土壤农杆菌转基因的抑菌抗生素 。
By a study of receptor system of heredity conversion in tissue culture, we have found out that, with MS as basic medium, BA0.2 + NAA2.0 as hormone compose and density, if using a 10-day sprout and taking the true leaf (Size 0.5 1.5 ㎝2) as outside plant, harming issue can form in two weeks.The study shows that, with MS+BA0.5 + NAA0.5 as medium, when adding 30g/L cane sugar and taking the true leaf as outside plant, we can have uncertain bud in two weeks.We have also discovered that, when using 75 g/ml Gent as the choice pressure of Agrobacterium tumerfaciens, 50 g/ml Gent as the choice pressure of turning gene plant and 300 g/ml cef as germ antibiotics, the result of heredity conversion on Erigeron Breviscapus (Vant) hand-mazz is the best.
