摘要 目的 探讨抗氧化剂维生素E(VitE)对氧化型低密度脂蛋白 (ox LDL)引起的人脐静脉内皮细胞 (HUVEC)毒性作用及增殖的影响。方法 以乳酸脱氢酶 (LDH)及3H 胸腺嘧啶 (3H TdR)、3H 亮氨酸 (3H Leu)掺入 ,分别测定HUVEC的损伤和增殖。结果 VitE可明显抑制高浓度 (>2 0 0 μg/L)ox LDL引起的内皮细胞释放乳酸脱氢酶(LDH) ,并能有效抑制低浓度 (<2 0 0 μg/L)ox LDL引起的HUVEC 3H TdR、3H Leu的掺入 ,二者均具有剂量依赖性。结论 抗氧化剂VitE对ox LDL引起的内皮细胞损伤有保护作用 ,对其引起的增殖有抑制作用 。 Objective To investigate the effect of antioxidant vitamin E on the cytotoxicity and proliferation of human umbilical vein endothelial cells (HUVEC) induced by ox-LDL.Methods The injury and proliferation of EC were determined respectively by lactic dehydrogenase (LDH) release kit and the incorporation of [ 3H]TdR and [ 3H]Leu into cells.Results VitE can significantly inhibit HUVEC to release LDH induced by high levels (>200 μg/L) of ox-LDL and the incorporation of 3 H-TdR and 3 H-Leu caused by low concentrations (<200 μg/L) of ox-LDL in dose-dependent manner.Conclusions Antioxidant VitE has both protective effect on the EC against injury and inhibitory effect on the proliferation of EC induced by ox-LDL.The results have great significance for preventing the formation of atherosclerosis and restenosis.
