摘要
目的 探讨异丙酚对大鼠肝脏缺氧复氧损伤中的保护作用。方法 将成年Wistar健康大鼠 2 4只随机均分为 3组 :对照组(A组 ) ,缺氧复氧组 (B组)和异丙酚预处理组 (C组 )。采用酶联免疫吸附法检测大鼠肝组织 8 异前列腺素F2α(8 iso PGF2α)水平 ,硝酸还原法测定其一氧化氮合酶 (NOS)活力和NO含量。结果 缺氧复氧后 ,B组肝组织中 8 iso PGF2α水平明显升高 ,而NOS活力和NO含量则明显降低 ,与A组比较差异有显著意义 (P <0 0 1)。C组与B组相比 ,肝组织中 8 iso PGF2α水平明显下降 ,而NOS活力和NO含量明显上升 ,差异亦有显著意义 (P <0 0 1)。结论 异丙酚能明显降低缺氧复氧后肝组织中的 8 iso PGF2α水平 ,提高内源性NO生成 ,提示对大鼠肝脏缺氧复氧损伤有明显的保护作用。
Objective To investigate the protective effects of propofol on liver against hypoxia/reoxgenation (H/R) injury in rats. Methods Twenty-four Wistar rats of both sexet weighing 200-250g were randomized into control group (group A), hypoxia/reoxygenation group (group B) and H/R + propofol group (group C). The 8-iso-prostaglandin F 2α (8-iso-PGF 2α ) level in the hepatic tissue was determined by enzyme-linked immunosorbent assay (ELISA). The content of NO and the activity of NOS were assessed by biochemical methods. Results In group B, the 8-iso-PGF 2α level in hepatic tissue was significantly higher than that in group A (P<0.01), and the content of NO and the activity of NOS were significantly lower than those in group A (P<0.01). The level of 8-iso-PGF 2α of group C decreased significantly and was lower than that of group B (P<0.01) . The content of NO and the activity of NOS of group C increased significantly and were lower than those of group B (P<0.01). Conclusions Propofol can decrease the level of 8-iso-PGF 2α and increase the content of NO. in hepatic tissue in an incident of hyponia/reoxygenation in rat, and it may inply that propofol may esert protective effect on the liver against such injury.
