摘要
目的研究高糖对人腹膜间皮细胞(HPMC)分泌纤维连接蛋白(FN)和血浆纤溶酶原激活物抑制剂-1(PAI-1)的影响,并进一步探讨川芎素对高糖致HPMC分泌FN和PAI-1的干预作用。方法采用胰蛋白酶消化法从人腹膜组织中分离间皮细胞,建立体外培养模型;用酶联免疫双抗夹心法(ELISA)检测HPMC培养液中FN和PAI-1蛋白质水平;用逆转录多聚酶链反应(RT-PCR)检测HPMC细胞内FN和PAI-1mRNA的表达。结果与单纯对照组相比,2.5%高糖刺激后,HPMC 培养液内FN和PAI-1蛋白质水平显著升高(P< 0.01),并上调HPMC FN和PAI-1的mRNA表达;加入低剂量(20,40μg/ml)川芎素后,FN和PAI-1蛋白质水平显著下降(P<0.01),mRNA表达水平下调。但随着剂量的加大,川芎素的上述作用减低乃至消失。结论高糖可引起HPMC FN和PAI-1蛋白质水平增加,并上调HPMC FN和PAI-1mRNA的表达,适当剂量的川芎素可拮抗高糖的上述作用。
To investigate the effect of sodium ferulate on high glucose-induced fibronectin (FN) and plasminogen activator inhibitor-1 (PAI-1) over expression in cultured human peritoneal mesothelial cells (HPMC). Mesothelial cells were isolated from human omental specimens by trypsin disaggregation and established a HPMC culture model. FN and PAI-1 protein in culture supernatants was measured by enzyme- linked immunosorbent assay (ELISA). Reverse transcriptase and polymerase chain reaction (RT-PCR) was used to detect the expression of FN messenger RNA (mRNA) and PAI-1 mRNA in HPMC. Compared with F12 group, The levels of FN and PAI-1 in culture supernatants increased significantly (P <0.01) and expression of FN mRNA and PAI-1 mRNA was up-regulateed in HPMC exposured to high glucose. These effects were reduced by sodium ferulate (P <0.01). [Conclusions] High glucose can increase the levels of FN and PAI-1 and up-regulate the expression of FN and PAI-1 mRNA. Sodium ferulate can reduce the effect of high glucose on HPMC.
