摘要
目的 :探讨川芎嗪对腹膜间皮细胞CD4 0表达的影响。方法 :分离、培养大鼠腹膜间皮细胞 ,分别用 4 2 5 %腹透液、4 2 5 %腹透液加γ 干扰素 (IFN γ ,10 0u/ml)、4 2 5 %腹透液加川芎嗪 4 0mg/L、4 2 5 %腹透液加IFN γ 10 0u/ml加川芎嗪 4 0mg/L刺激细胞 30min ,以DMEM /F12培养基为对照组。通过逆转录 多聚酶链反应 (RT PCR)及流式细胞仪 (FACS)检测间皮细胞CD4 0表达。结果 :4 2 5 %腹透液刺激后腹膜间皮细胞CD4 0mRNA及其蛋白的表达明显高于对照组 (P <0 0 1) ;4 2 5 %腹透液加IFN γ联合刺激后 ,间皮细胞CD4 0mRNA及其蛋白的表达明显高于对照组及单纯 4 2 5 %腹透液组 (P <0 0 1)。加入川芎嗪后 ,可显著减低单纯 4 2 5 %腹透液及 4 2 5 %腹透液加IFN γ引起的CD4 0mRNA及其蛋白的高表达 (P <0 0 1)。结论 :单纯腹透液及腹透液加IFN γ可显著增加腹膜间皮细胞CD4 0的表达 ,而川芎嗪对腹膜间皮细胞CD4 0的表达有显著的抑制作用。在长程腹膜透析 (CAPD)过程中 ,加入川芎嗪可能有助于减轻透析液引起的慢性炎症反应 ,从而防止或延缓腹膜纤维化的发生。
Objective: To explore the effect of Ligustrazine (LGZ) affecting the expression of CD40 on mesothelial cells (MC). Methods: Rat′s peritoneal MC isolated from peritoneal cavity were cultured and maintained under the defined conditions in vitro. They were cultured with 4.25% peritoneal dialysis solution (PDS), 4.25% PDS added LGZ (40mg/L), 4.25% PDS added IFN γ (100u/ml) and 4.25% PDS added IFN γ and LGZ for 30 min, the DMEM/F12 cultural medium was taken as control. The expression of CD40 on MC was detected by reverse transcript polymerase chain reaction (RT PCR) and flow cytometry analysis. Results: MC cultured in vitro expressed CD40 markedly, the expression of CD40 mRNA and its protein was markedly up regulated following stimulation with 4.25% PDS or IFN γ and 4.25% PDS. LGZ (40mg/L) could reduce the expression of CD40 mRNA and its protein significantly as it was added in 4.25% PDS and 4.25% PDS with IFN γ. Conclusion: The PDS stimulates the expression of CD40 on MC. LGZ has the function of down regulating the expression of CD40 on MC. During long term continuous ambulatory peritoneal dialysis, it might be helpful to alleviate chronic inflammatory reaction by adding LGZ into standard PDS, therefore prevent or retard the occurence and development of peritoneal fibrosis.
