摘要
作者采用SDS-聚丙烯酰胺凝胶电泳和免疫印迹技术,对黄疸出血群赖型017株和601株、七日热群七日热型156株、澳洲群澳洲型620株以及塞马伦群帕托克型Patoc Ⅰ株等5株钩体外膜进行分析。检测到具有免疫保护作用和抑制钩体粘附作用的单克隆抗体E_4B_7D_5能特异地识别赖型017株和601株钩体外膜的34.5kd和39.5kd两条蛋白带,而同其余3株钩体外膜蛋白无反应。由此提示该两条蛋白带可能为钩体外膜保护性抗原。
McAb E4B7D5 was prepared by hybridoma technology in BALB/c mice immunized to outer envelope of Lep-tospira interrogans serovar lai. This McAbagglutinated rpecifically with all the 13serovars of Icterohaemorrhagiae serogroupin MAT test at high titres and protectedthe guinea pigs against the attack of viru-lent strain (017) of serovar lai. SDS-PAGEand immunoblot were used to analyse thereaction of the outer envelopes of the fivestrains of Leptospira (Leptospira interrogansIcterohaemorrhagic serogroup, serovar lai,017 and 601 stains; L.interrogans Hebdom-adis serogroup, hebdomadis serovar strain156; L. interragans Australis serogroup aus-tralis serovar strain 620; biflexa leptospirapatoc serovar strain Patoc I; with McAbE4B7D5. Results indicated that this McAbE4B7D5 recognized specifically 34.5kd and39.5kd outer envelope proteins of serovarlai, strain 017 and 601, so it might be animmunoprotective McAb directed againstLeptospira interrogans serovar lai. It wassuggested that 34.5kd and 39.5kd outerenvelope proteins of serovar, lai recognizedspecifically by McAb E4B7D5 might beimmunoprotective antigens.
