摘要
目的探讨支气管哮喘患者 β2 肾上腺素能受体 (β2 AR) 16、2 7位点基因多态性与血清总IgE的相关性。方法采用等位基因特异性聚合酶链反应 (AS PCR)方法 ,对 4 4例支气管哮喘患者进行 β2 AR基因多态性分析 ,并测定其血清总IgE。结果各基因型血清总IgE的值 ,16位点 :精氨酸 /精氨酸为 5 2 5 2 5± 2 2 0 73mmol/L ,甘氨酸 /精氨酸为 4 19 87± 2 6 4 0 8mmol/L ,甘氨酸 /甘氨酸为 5 35 10±2 89 5 1mmol/L ,三者比较差异无显著性 (P >0 0 5 ) ;2 7位点 :谷氨酰胺 /谷氨酰胺为 75 6 13±2 2 0 6 3mmol/L ,谷氨酸 /谷氨酰胺为 341 0 0± 136 6 6mmol/L ,谷氨酸 /谷氨酸为 4 2 5 33± 2 74 13mmol/L ;三者比较差异有显著性 (P <0 0 1)。结论 β2 AR基因 2 7位点多态性与血清总IgE相关联。
Objective To analyze the association between β 2-AR genetic polymorphism and serum total IgE in asthma patients. Methods AS-PCR were used to determine 16, 27 locus alleles of β 2-AR genetic polymorphisms and ELISA were used to determine serum total IgE in 44 unrelated patients with asthma. Results The relativity between serum total IgE and genotype β 2-AR16 loci: the level of IgE was 525.25±220.73mmol/l in Arg/Arg, 419.87±264.08mmol/l in Arg/Gly and 535.10± 289.51 mmol/l in Gly/Gly respectively. There was no significant difference in serum total IgE compared with three groups. The level of IgE were 756.13±220.63mmol/l in Gln/Glu, 341.00±136.66mmol/l in Gln/Glu and 425.33±274.13mmol/l in Glu/Glu respectively. There were significant difference in β 2-AR 27 locus of serum total IgE compared with three groups. Conclusion β 2-AR 27 locus genetic polymorphism is correlated with serum total IgE in asthma patients.
