摘要
全乙酰化的表没食子儿茶素没食子酸酯在酸性条件下水解可以得到表没食子儿茶素没食子酸酯(EGCG),但此反应随时间的延长伴随有聚合、氧化等副反应。为得到纯的表没食子儿茶素没食子酸酯,优化水解条件,用高效液相跟踪反应,选甲醇和水(体积比为90∶10)为流动相,检测器紫外波长为230 nm,每30 min检测1次,最后确定水解反应在270min完成。以较高收率得到了高纯度的表没食子儿茶素没食子酸酯,并经熔点、比旋光度、核磁共振氢谱、碳谱和高分辨质谱确证。
Acidic hydrolysis of epigallocatechin-3-gallate peracetate provided Epigallocatechin-3-gallate.To avoid the polymerization and oxidation in the hydrolysis process,the reaction was traced by HPLC every 30 minutes.The mobile phase was methanol and water(90/10,V/V),and the detector wavelength was 230 nm.The hydrolysis reaction finished after 270 minutes.Under this hydrolysis condition,epigallocatechin-3-gallate was prepared and confirmed by melting point,optical rotation,1HNMR,13C NMR and HRMS.
