摘要
目的:研究不同剂量消癌平对白血病细胞HL-60黏附、运动和侵袭的影响。方法:HL-60细胞体外培养,经0、0.1、0.15、0.5g/L消癌平分别处理细胞,细胞黏附实验检测细胞黏附力,细胞迁移实验检测细胞运动力,细胞侵袭实验检测细胞侵袭力,RT-PCR检测HL-60细胞MMP-2和MMP-9 mRNA的表达。结果:与对照组比较,0.1、0.1 5、0.2g/L消癌平不仅能有效抑制HL-60细胞的黏附力、运动力和侵袭力(P<0.01),而且还能下调HL-60细胞MMP-2和MMP-9mRNA的表达(P<0.0 1)。结论:消癌平能抑制HL-60的浸润转移能力,其机制可能与下调MMP-2和MMP-9 mRNA的表达使HL-60细胞的黏附力、运动力和侵袭力下降有关。
AIM:To study the inhibitory effects of TXYP on adhesion,mingration and invasion of leukemia cells HL-60 METHODS:HL-60 cells were cultured in vitro and treated by 0,0.1,0.15,0.2 g/L TXYP:Then cell adhesion assay,cell migration assay and cell invasion assay were used to observe the effects of matrine on adhesion,migration and invasion of HL-60 cells,and RT-PCR were performed to evaluate the MMP-2 and MMP-9mRNA expression levels.RESULTS:Contrary to the control group, 0.1、0.15、0.2g/L TXYP could not only effectively inhibit the adhesion,migration and invasion of HL- 60 cells(P<0.01).CONCLUSION:By way of down-regulating the expressions of MMP-2和MMP-9 mRNA,TXYP can inhibit the capability of adhesion,migration and invasion of 60 cells.
