摘要
目的研究不同浓度灵芝三萜组分GLE对人鼻咽癌细胞CNE2侵袭转移的影响及其作用机制。方法 CNE2细胞体外培养,经10μg/mL、15μg/mL和20μg/mL GLE分别处理1h、24h及48h,细胞黏附实验检测细胞黏附能力,细胞迁移实验检测细胞运动能力,细胞侵袭实验检测细胞侵袭能力。应用蛋白免疫印迹法检测侵袭转移相关蛋白的表达。结果不同浓度GLE与对照组比较,GLE能有效抑制CNE2细胞的黏附力、运动力和侵袭力(P<0.05),且能下调CNE2细胞中MMP-2和MMP-9蛋白的表达。结论 GLE抑制CNE2细胞的浸润转移能力,其机制可能与下调MMP-2和MMP-9蛋白的表达使CNE2细胞的黏附力、运动力和侵袭力下降有关。
Objective To study the effects of Triterpene GLE from Ganederma Lucidum on invasion and metastasis of CNE2 cells. Methods CNE2 cells were cultured in vitro and treated by 10μg/mL, 15μg/mL and 20μ g/mL GLE. Then cell adhesion assay,cell migration assay and cell invasion assay were used to observe the effects of GLE on adhesion, migration and invasion of CNE2 cells. Western blotting was used to detect invation and metastasisassociated protein level. Results Contrasting to control group. 10 μg/mL, 15μg/mL and 20μg/mL GLE could not only effectively inhibit the adhesion, migration and invasion of CNE2 cells ( P 〈 0.05 ), but also down - regulate the expression of MMP - 2 and MMP - 9. Conclusion By down - regulating the expressions of MMP - 2 and MMP - 9, GLE could inhibit the capability of adhesion,migration and invasion of CNE2 cells.
