摘要
本实验旨在合成和鉴定双烯雌酚(DEN)免疫抗原,通过双烯雌酚的酚羟基与琥珀酸酐反应,合成半抗原双烯雌酚-琥珀酰半酯(DEN-HS),采用混合酸酐法将其与载体蛋白BSA偶联制备免疫抗原(DEN-BSA)经紫外光谱分析和动物免疫试验证实免疫抗原合成成功,间接酶联免疫法分析证明其免疫鼠所获得的抗体与双烯雌酚有良好的特异性。实验成功合成了双烯雌酚免疫抗原,并获得了高效价的免疫血清,研究结果为免疫分析方法的进一步研究提供了试验基础。
To synthesize and identify artificial antigen of dienestrol,the artificial antigen of dienestrol( DEN-BSA) synthesized through the reaction between succinate acid anhydride and dienestrol were coupled with bovine serum albumin(BSA) by the mixed acid anhydride reaction.The results from UV,SDS-PAGE showed that DEN-BSA had been conjugated successfully.The artificial antigens were synthesized successfully and the anti-dienestrol antibody received.This study provided the basis for further research to establish immunoassay of dienestrol.
