摘要
目的 探讨大鼠脊髓损伤后两种结构型一氧化氮合酶( cNOS) mRNA表达的变化。方法 以 Nystrom方法建立大鼠脊髓压迫伤模型,用逆转录聚合酶链反应( RT- PCR)法测定伤段脊髓组织神经型及诱导型 NOS( nNOS, eNOS) mRNA的表达情况。结果 脊髓压迫伤后 nNOS mRNA及 eNOS mRNA表达均增强,伤后 6 h达到高峰, eNOS mRNA表达增强更明显。结论 脊髓损伤后结构型 NOS mRNA的表达主要在伤后早期增强,不同的结构型 NOS mRNA表达增强的程度不同。
Objective To study the changes of constructive nitric oxide synthase (cNOS) gene expression in injured spinal cord tissue. Method Establish a compression injury model of rat spinal cord and gene expression levels of neuronal NOS (nNOS) and endothelial NOS (eNOS) are measured by reverse transcription- polymerase chain reaction (RT- PCR).Result Expression of nNOS mRNA and eNOS mRNA were upregulated after injury,reaching the maximum 6 h after injury, while increase of the expression of eNOS mRNA was especially significant. Conclusion cNOS gene expression increased,especially in early stage, after spinal cord injury , and expression of eNOS mRNA is higher than that of nNOS mRNA.
