摘要
目的:探讨梗阻性黄疸大鼠外周血中性粒细胞凋亡的改变。方法:将66只成年Wistar大鼠随机分为正常(A)组、假手术(B)组和胆总管结扎(CBDL,C)组,B、C组术后又分为1、3、7、10、14d5个时相点;应用流式细胞仪检测中性粒细胞(PMN)的凋亡指数(AI),通过ELISA双抗体夹心法测定血清IL-8。结果:C组大鼠术后各时相点PMN的AI显著降低,与A组和B组均有显著差异(P<0.05);C组大鼠术后各时相点血清IL-8水平显著升高,与A组和B组均有显著差异(P<0.05);且血清IL-8水平与PMN的AI的改变有显著负相关性。结论:梗阻性黄疸大鼠外周血PMN的AI明显降低,且与IL-8水平异常升高有关,在梗阻性黄疸过度炎症反应的发生、发展中有着重要意义。
Objective To investigate the changes of apoptosis of peripheral polymorphonuclear neutrophil (PMN) in the rats with obstructive jaundice. Methods 66 Wistar adult rats were randomly divided into three groups: normal group (group A), sham operation group (group B) and common bile duct ligation group (CBDL, group C), and each group were randomly divided to subgroups of day 1, 3, 7, 10, 14 again. The flow cytometry was used to detect and evaluate the change of apoptosis in vitro after PMN were harvested from the rats. IL-8 was detected by ELISA method. Results Apoptosis index (AI) of PMN in group C was lower than in groups A and B at 1, 3, 7, 10, 14 days after CBDL; The level of IL-8 in serum in group C was higher than in groups A and B at 1, 3, 7, 10, 14 days after CBDL. Moreover, there was a marked negative correlation between AI and level of IL-8. Conclusion The apoptotic PMN is decreased, which is intimately associated with the level of IL-8, which may induce excessive inflammation, and play a crucial role in the initiation and development of obstructive jaundice.
