摘要
目的观察阿那其根醇提取物(EEAP)对咳嗽变异性哮喘(CVA)豚鼠脑组织炎症因子和氧化指标的影响。方法将48只雄性豚鼠随机分为6组,分别为空白对照组、CVA模型组、EEAP低剂量组(160 mg/kg)、EEAP中剂量组(320 mg/kg)、EEAP高剂量组(640mg/kg)和阳性对照组(醋酸泼尼松,250mg/kg),每组8只。除空白对照组外,其余各组豚鼠均皮下注射1 mg/mL卵清蛋白(OVA)-费氏完全佐剂溶液致敏,并雾化吸入1%OVA-生理盐水溶液以激发哮喘。末次雾化后,各给药组大鼠均灌胃相应药物,对照组和模型组大鼠均灌胃等体积水。雾化吸入10-4mol/L的辣椒素溶液诱导各组豚鼠咳嗽,记录咳嗽次数。ELISA法检测各组豚鼠脑组织肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)、超氧化物歧化酶(SOD)和丙二醛(MDA)水平。结果1.与空白组相比,CVA模型组豚鼠咳嗽次数显著升高,差异具有统计学意义(P<0.001);与CVA模型组相比,低、中、高剂量EEAP组和阳性对照组豚鼠咳嗽次数明显减少,差异有统计学意义(P<0.01,P<0.001,P<0.001,P<0.001)。2.与空白对照组相比,CVA模型组豚鼠脑组织TNF-α和IL-6水平升高,差异具有统计学意义(P<0.05,P<0.05);与CVA模型组相比,高剂量EEAP组和泼尼松组豚鼠脑组织TNF-α和IL-6水平下降,差异具有统计学意义(P<0.05,P<0.05)。3.与空白对照组相比,CVA模型组豚鼠脑组织SOD和MDA水平无差异,无统计学意义(P>0.05,P>0.05)。4.空白对照组与CVA模型组豚鼠咳嗽次数与脑组织TNF-α和IL-6水平呈正相关(R=0.658,P=0.006;R=0.623,P=0.01)。结论EEAP具有降低咳嗽变异性哮喘豚鼠脑组织TNF-α和IL-6水平,改善大脑炎性,但其作用机制仍需进一步研究。
Objective To observe the effects of pyrethrum root alcohol extract(EEAP)on inflammatory factors and oxidation indexes in the brain tissue of guinea pigs with cough variant asthma(CVA).Methods 48 male guinea pigs were randomly divided into 6 groups:the blank control group,the CVA model group,the EEAP low dose group(160 mg/kg),the EEAP medium dose group(320 mg/kg),the EEAP high dose group(640 mg/kg)and the positive control group(prednisone acetate,250 mg/kg),8 rats in each group.Except the blank control group,the other groups of guinea pigs were subcutaneously injected with 1 mg/m L ovalbumin(OVA)-Fischer’s complete adjuvant solution to sensitize and inhaled 1%OVA-physiological saline solution to stimulate asthma.After the last atomization,the rats in each group was given the corresponding drugs,and the rats in the control group and the model group were given the same volume of water.Inhalation of a 10-4 mol/L capsaicin solution induced a cough in each group of guinea pigs,and the number of coughs were recorded.The levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),superoxide dismutase(SOD)and malondialdehyde(MDA)in guinea pig brain tissues were detected by ELISA.Results 1.Compared with the blank group,the cough frequency of guinea pigs in the CVA model group was significantly increased,and the difference was statistically significant(P<0.001);compared with the CVA model group,the number of coughs in guinea pigs in the low,medium,and high dose EEAP groups and the positive control group was significantly reduced,and the differences were statistically significant(P<0.01,P<0.001,P<0.001,P<0.001).2.Compared with the blank control group,the levels of TNF-αand IL-6 in the brain tissue of guinea pigs in the CVA model group were increased,and the difference was statistically significant(P<0.05,P<0.05);compared with the CVA model group,the dose was higher The levels of TNF-αand IL-6 in the brain tissue of guinea pigs in EEAP group and prednisone group decreased,and the difference was statistically significan
