Optimization of Extraction Process of Qingyan Zhitong Prescription Based on Spherical Symmetry Design and GA-BP Model Combined with AHP-CRITIC

摘要 目的:采用球面对称设计和遗传-神经网络(GA-BP)模型优选清咽止痛方的提取工艺。方法:采用球面对称设计结合层次分析法(AHP)、基于指标相关性的权重确定方法(CRITIC)及AHP-CRITIC混合加权法,以浸泡时间、加水倍数及煎煮时间为考察因素,以多指标成分(绿原酸、连翘苷、连翘酯苷A、牛蒡子苷、迷迭香酸、黄芩苷与甘草苷)含量、干膏率及指纹图谱相似度为综合评价指标,并结合GA-BP模型优选清咽止痛方的最佳提取工艺,并进行工艺验证。结果:根据球面对称设计试验结果,建立了结构为3-4-1的三层BP人工神经网络,结合遗传算法筛选出最优提取工艺为:浸泡120 min、加17倍量水、煎煮3次、每次30 min,该条件下的综合评分均值为87.2555。结论:球面对称设计结合GA-BP模型可用于优选清咽止痛方的提取工艺,为清咽止痛方的工业化生产提供了依据。 Objective:To optimize the extraction process of Qingyan zhitong prescription based on spherical symmetry design and genetic neural network(GA-BP)model.Methods:Using spherical symmetric design combined with analytic hierarchy process(AHP),weight determination method based on index correlation(CRITIC)and AHP-CRITIC mixed weighting method,soak time,water ratio and extraction time were used as factors,multi-index constituents(chlorogenic acid,forsythin,forsythoside,gastrodin,rosmarinic acid,baicalin and liquiritin)contents,dry extract rate and fingerprint similarity were used as the comprehensive evaluation index,and combined with GA-BP model,the extraction process of Qingyan zhitong prescription was optimized,process validation was carried out.Results:According to the experimental results of spherical symmetry design,a three-layer BP artificial neural network with the structure of 3-4-1 was established.Combined with genetic algorithm,the optimal extraction process was soaking 120 min,adding 17 times of water,decocting 3 times,30 min each time.The average comprehensive score under this condition was 87.2555.Conclusion:Spherical symmetry design combined with GA-BP model can be used to optimize the extraction process of Qingyan zhitong prescription,which provides a basis for the industrial production of Qingyan zhitong prescription.
