The plasma membrane Na+/H+-antiporter salt overly sensitive1 (SOS1) from the halophytic Arabidopsis-relative Thellungiella halophila (ThSOS1) shows conserved sequence and domain structure with the orthologous genes from Arabidopsis thaliana and other plants. When expression of ThSOSt was reduced by RNA interference (RNAi), pronounced characteristics of salt-sensitivity were observed. We were interested in monitoring altered transcriptional responses between Thellungiella wild type and thsost-4, a representative RNAi line with particular emphasis on root responses to salt stress at 350 mmol/L NaCI, a concentration that is only moderately stressful for mature wild type plants. Transcript profiling revealed several functional categories of genes that were differently affected in wild-type and RNAi plants. Down-regulation of SOS1 resulted in different gene expression even in the absence of stress. The pattern of gene induction in the RNAi plant under salt stress was similar to that of glycophytic Arabidopsis rather than that of wild type Thellungiella. The RNAi plants failed to down-regulate functions that are normally reduced in wild type Thellungiella upon stress and did not up-regulate functions that characterize the Thellungiella salt stress response. Metabolite changes observed in wild type Thellungiella after salt stress were less pronounced or absent in RNAi plants. Transcript and metabolite behavior suggested SOS1 functions including but also extending its established function as a sodium transporter. The down-regulation of ThSOS1 converted the halophyte Thellungiella into a salt-sensitive plant. 展开更多

An analytical solution in the Laplace space for the pressure-transient behavior for a line-source, constant-rate well of this type of reservoir is obtained with the Laplace transformation and the perturbation technique. The pressure and its derivative in the reservoir and the effects of relevant parameters on the pressure-transient response are obtained. The model as well as the corresponding type curves may be used in predicting the production performance or analyzing the production data for this type of reservoir. 展开更多

Analyzing the potential mechanism of Buyang Huanwu decoction for the treatment of salt-sensitive hypertension based on network pharmacology and in vivo experiments

Background:Buyang Huanwu decoction(BHD)is a traditional Chinese medicine herbal formula used for treating hypertension,particularly in the later stages of hypertension when it is associated with intracerebral hemorrhage.This study aims to investigate the treatment mechanism of BHD to provide a basis for its clinical application in hypertension treatment.Methods:Network pharmacology analysis and cell culture experiments were performed to explore the potential proteins and mechanisms of action of BHD against hypertension.Bioactive compounds related to BHD were screened,and relevant targets associated with hypertension and BHD were retrieved.Molecular docking technology was used to identify the effective signaling pathway based on the Kyoto Encyclopedia of Genes and Genomes and protein-protein interaction network cores.Lastly,the effects and mechanisms of BHD on salt-sensitive hypertensive endothelial cells were investigated.Results:Ninety-three potential therapeutic targets for BHD and salt-sensitive hypertension were found to be closely associated with the PI3K/Akt/eNOS signaling pathway and oxidative stress.Cell experiments further indicated the pivotal role of endothelial cells in hypertension,and validation analysis showed that BHD significantly preserved cell morphology,suppressed oxidative stress reactions,activated the PI3K/Akt/eNOS signaling pathways,preserved normal endothelial cell function,and reduced cell apoptosis.Conclusion:BHD effectively activates the PI3K/Akt/VEGF signaling pathway,attenuates oxidative stress-induced injury in endothelial cells exposed to high salt levels,and mitigates apoptosis,supporting the use of traditional Chinese medicine BHD in the treatment of salt-sensitive hypertension. 展开更多

In atherosclerosis,chronic inflammatory processes in local diseased areas may lead to the accumulation of reactive oxygen species(ROS).In this study,we devised a highly sensitive H_(2)O_(2)-scavenging nanobionic system loaded with probucol(RPP-PU),to treat atherosclerosis more effectively.The RPP material had high sensitivity to H_(2)O_(2),and the response sensitivity could be reduced from 40 to 10μmol/L which was close to the lowest concentration of H_(2)O_(2)levels of the pathological environment.RPP-PU delayed the release and prolonged the duration of PU in vivo.In Apolipoprotein E deficient(ApoE-/-)mice,RPP-PU effectively eliminated pathological ROS,reduced the level of lipids and related metabolic enzymes,and significantly decreased the area of vascular plaques and fibers.Our study demonstrated that the H_(2)O_(2)-scavenging nanobionic system could scavenge the abundant ROS in the atherosclerosis lesion,thereby reducing the oxidative stress for treating atherosclerosis and thus achieve the therapeutic goals with atherosclerosis more desirably. 展开更多

The advanced temperature and stress test machine was introduced to determine the early cracking tendency of concrete with inclusion of light-burnt MgO under full restraint by tracking the development of thermal,physical and deformation properties.Results showed that light-burnt MgO being incorporated ranging between 4 wt% and 6 wt% of cementitious materials was beneficial to increase the maximum compressive stress and cracking stress of concrete by 0.37 MPa and 0.2 MPa on average respectively.The second zero stress temperature was reduced by 11.4 ℃ and the maximum temperature was slightly reduced while cracking thermal impact was significantly enhanced from 59.8 ℃ to 66.2 ℃.Sensitive anti-cracking coefficient F was forwarded to assess the early cracking tendency of concrete and the inclusion of 4 wt% light-burnt MgO with activity of 109 s ranked the best in crack resistance. 展开更多

支承运转件(顶枢、底枢、蘑菇头等)是船闸闸门的重要组成部分,也是闸门系统的典型薄弱部位,频繁的闸门启闭及水流紊动等情况会令支承运转件出现磨损、卡阻等运行故障,由于部分支承运转件处于水下隐蔽位置,通过现有直接检测方法难以评估... 支承运转件(顶枢、底枢、蘑菇头等)是船闸闸门的重要组成部分,也是闸门系统的典型薄弱部位,频繁的闸门启闭及水流紊动等情况会令支承运转件出现磨损、卡阻等运行故障,由于部分支承运转件处于水下隐蔽位置,通过现有直接检测方法难以评估其受力特性及磨损程度,研究闸门及支承运转件敏感区的应力关系对支承运转件的运行安全具有重要意义。为此,文章依托某实际船闸工程项目,建立船闸闸门及其支承运转件的整体有限元模型,基于数值模拟仿真分析方法,研究了在不同水位、不同闸门开度等工况下船闸闸门及其支承运转件的应力响应规律,确定了门叶和支承运转件的应力敏感区域,定量分析了船闸闸门与支承运转件敏感区的应力响应关系,实现了通过闸门受力情况来初步判别其支承运转件受荷特征的目的,从而间接评测闸门支承运转件的运行故障。研究成果可为运行状态下船闸闸门的智能监测、安全预警等提供技术支撑。 展开更多

The Impact of Minerals on Wheat Plants Grown under Salinity Stress

The present work has been conducted to study the impact of Zn (20 and 200 μM) or Ni (1 and 100 μM) on growth, metabolic activities, osmotic pressure and heavy metal of wheat cultivar (cv. Gimiza 11) under salt stress conditions. Harvest index was significantly decreased by 75% at -0.3 MPa and -1.2 MPa decreased by 53%. Length of root, stem and spike, and leaf area were slightly reduced up to -0.9 MPa NaCl level, after that a significant reduction was recorded. Total sugar was increased in the shoot and spike, while decreased in the root with increasing salinity levels. On the other side, total protein was increased in the shoot while decreased in both the root and spike. Osmotic pressure was increased as increasing salinity levels. Treatment of wheat plants with Zn or Ni alleviated generally salt injury in the harvest index, length of root, shoot and spike and leaf area. This effect was more obvious with Zn treatments. Mineralization treatments increased total sugar in the three tested organs while did not change the total protein. Zn content was increased at -1.2 MPa level while Ni was decreased. Treatments with Zn or Ni mostly increased Zn while causing a reduction in the Ni content. These treatments also increased the values of osmotic pressure compared with the equivalent levels of control plants. 展开更多

The purpose of the present paper is to modify and extend the surface shear stress measurement technique de-termined by Reda et al. to wind tunnel studies. All the facilities employed in the technique are very common and simple. The measurement technique is applied to surface flow beneath a tangential jet and surface flow in a turbulence wedge generated by a small cylinder in windtunnel flow. Recent studies revealed that DNA methylation plays an important role in plant growth and development. In this study, a water-saving and drought-resistant rice variety Huhan 3 was subjected to drought stress from tillering to grain-filling stages in six successive growth cycles. The variations in DNA methylation pattern between the original generation (Go) and the sixth generation (G6) were analyzed by using methylation sensitive amplification polymorphism method. The results revealed that the methylated loci accounted for 34.3% to 34.8% of the total loci. Among these methylated loci, 83.1% to 84.8% were full- and hyper-methylated and 15.2% to 16.9% were hemi-methylated. The DNA methylation level decreased from the three-leaf to four-leaf stages in Huhan 3. Differentially methylated loci (DML) between generations or/and between different developmental stages accounted for 4.0% of the total loci, most of which were only related to plant development (57.9%). Compared to Go, the DNA methylation pattern of G8 changed after drought domestication, at the three-leaf stage, de-methylation accounting for 59.1%, while at the four-leaf stage, re-methylation for 47.9%. Genome-wide alternations of DNA methylation were observed between the two seedling stages, and DML mainly occurred on the gene's promoter and exon region. 目的研究薄膜体声波滤波器的加速寿命评估技术,针对XXX1001型薄膜体声波滤波器进行可靠性寿命评估。方法根据薄膜体声波滤波器设计、结构特点,针对XXX1001型薄膜体声波滤波器典型应用环境条件下的失效模式和失效机理分析,采用环境影响... 目的研究薄膜体声波滤波器的加速寿命评估技术,针对XXX1001型薄膜体声波滤波器进行可靠性寿命评估。方法根据薄膜体声波滤波器设计、结构特点,针对XXX1001型薄膜体声波滤波器典型应用环境条件下的失效模式和失效机理分析,采用环境影响分析明确寿命表征指标和相应敏感应力,以此确定合适的加速寿命方法和加速模型。结合可靠性摸底试验确定的敏感应力极限和加速模型要求,设计加速寿命试验方案,利用阿伦尼斯模型对寿命表征指标等试验数据进行评估,得到该型薄膜体声波滤波器的可靠性寿命参数。结果确定了滤波器的寿命表征指标(插入损耗)和敏感应力(温度),设计了加速寿命试验方案,试验实施后,根据试验数据评估出XXX1001型薄膜体声波滤波器在40℃工作时的平均寿命为117 803h。结论该技术程序较为简单、实施方便、样本少、成本低,可为类似滤波器的可靠性评估提供参考。其评估结果也可为XXX1001型薄膜体声波滤波器的设计改进和应用推广提供技术支撑。 展开更多
