目的研究蒲公英多糖对溃疡性结肠炎合并菌群失调小鼠肠道菌群变化及抗炎作用。方法选择ICR清洁级小鼠70只,鼠龄4~8周,体质量19~23 g。在建立小鼠溃疡性结肠炎模型的基础上,采用林可霉素灌胃法诱导肠道菌群失调。将小鼠分为正常组、模型...目的研究蒲公英多糖对溃疡性结肠炎合并菌群失调小鼠肠道菌群变化及抗炎作用。方法选择ICR清洁级小鼠70只,鼠龄4~8周,体质量19~23 g。在建立小鼠溃疡性结肠炎模型的基础上,采用林可霉素灌胃法诱导肠道菌群失调。将小鼠分为正常组、模型组、阳性对照组和治疗组,各组随机分配10只。观察蒲公英多糖对小鼠一般状况、体质量、肠道菌群的影响;采用酶联免疫吸附分析法(ELISA)观察血清中尿酸(UA)、一氧化氮(NO)含量及血清中白细胞介素4(IL-4)、白细胞介素6(IL-6)、肿瘤坏死因子-α(TNF-α)的表达。结果正常组小鼠精神佳,体毛顺滑,进食、进水及二便均正常;模型组小鼠第2天开始出现精神倦怠乏力、行动迟缓、体毛毛躁、进食量减少、粪便稀糊不成型等状况。经过治疗后,阳性对照组和治疗组小鼠精神好转明显,体毛较前顺滑,活动量增加,摄食饮水量也有所增加,粪便逐渐成形。经蒲公英多糖治疗后,与模型组比较,治疗组小鼠一般状况改善,体质量增加(第5天:25.26 g±0.33 g vs 23.53 g±0.71 g;第7天:26.63 g±0.99 g vs 24.98 g±0.80 g。P<0.05);第7天,血清中UA、NO含量降低(77.94μmol/L±5.06μmol/L vs91.84μmol/L±9.35μmol/L,32.06μmol/L±4.43μmol/L vs 56.76μmol/L±4.24μmol/L。P<0.05);血清中IL-4升高(76.29 ng/mL±6.36 ng/mL vs 52.08 ng/mL±7.14 ng/mL。P<0.05),IL-6和TNF-α含量降低(IL-6:124.31 ng/mL±9.66 ng/mL vs 134.49 ng/mL±13.71 ng/mL;TNF-α:107.47 ng/mL±8.23 ng/mL vs 112.46 ng/mL±10.30 ng/mL。P<0.05);小鼠肠道内双歧杆菌和乳酸杆菌数量增加,大肠杆菌数量减少(P<0.05)。结论蒲公英多糖能够抑制溃疡性结肠炎合并菌群失调小鼠血清中UA、NO的产生,减少炎性因子的表达,调节正常菌群和益生菌数量,对改善林可霉素致小鼠肠道菌群失调有明显的作用。展开更多

Flavonoids were extracted from dandelion using alcohol precipitation ultrasonic extraction method and polysaccharide was extracted using water ul- trasonic extraction method. DPPH free radical scavenging ability of flavonoid and polysaccharide was determined using ultravialet spectrophotometer method. The difference in DPPH free radical scavenging ability between flavonoids and polysacchafide was analyzed statistically. The results showed that DPPH scavenging ability was the best when the concentration of flavanoid was 1.6 mg/mL; the scavenge rate was up to 96.11% and I Cso (median inhibitory concentration of inhibi^rs) was 0.38 mg/mL. DPPH scavenging ability was the best when the concentration of polysaccheride was 1.4 mg/mL; the scavenge rate was 89.33% and IC5o was 0.51 mg/mL. Statistical results showed that there were significant differences in DPPH free radical scavenging ability between dandelion flavonoid and polysaeoharide (P 〈 0.05 ). Flavonoid had significantly higher scavenging ability than polysaccharide (P 〈 0.05). The study lays a theoretical foundation for further research on dan- delion flavonoid, and provides a theoretical basis for development and utilization of dandelion resources.展开更多
