摘要
以新疆野生蒲公英为原料,通过超声辅助法提取蒲公英多糖后以不同体积分数的乙醇溶液(20%,40%,60%和80%)进行分级沉淀,依次得到4种蒲公英多糖样品(PD-20,PD-40,PD-60和PD-80);为探究不同组分间的差异,分别测定其总糖含量、蛋白质含量、糖醛酸含量及单糖组成,并通过紫外和红外光谱进一步分析不同组分之间的特性差异。结果表明,蒲公英多糖4个组分均为结合蛋白的复合物,其中PD-80组分的总糖含量最高(PD-80>PD-40>PD-60>PD-20),蒲公英多糖PD-20组分的蛋白质含量最高;单糖组成测定结果显示4种多糖样品均由阿拉伯糖、鼠李糖、甘露糖、葡萄糖和半乳糖组成,但摩尔比存在一定差异;4种多糖样品均具有一定的降血糖和抗氧化活性,且呈量-效关系。
With wild dandelion from Xinjiang as the raw material,dandelion polysaccharides which were extracted with ultrasound-assisted method were fractionated by different gradient concentrations of ethanol(20%,40%,60%,80%)into four precipitation fractions.To explore the differences between different components,the contents of total sugar,protein,galacturonic acid and monosaccharide composition were determined,UV and IR spectroscopy analysis were run simultaneously to determine their spectroscopy properties.The results indicated that all four fractions were combined with a certain amount of protein and the samples precipitated by 80%ethanol had higher sugar content,the samples precipitated by 20%ethanol had the highest protein content.All four fractions were composed of arabinose,rhamnose,mannose,glucose and galactose,but their molar ratio of monosaccharide had significant differences.All four fractions had certain hypoglycemic antioxidant activity,and its hypoglycemic antioxidant effect showed a good dose-effect relationship with the concentration of polysaccharides.
