蒲公英多糖对幽门螺杆菌相关性胃炎大鼠胃黏膜炎性反应及MAPK/ERK通路的影响 被引量:49

Effect of dandelion polysaccharide on gastric mucosal inflammatory response and MAPK/ERK pathway in rats with Helicobacter pylori-associated gastritis

摘要 目的从抗炎、调节MAPK/ERK通路方面探讨蒲公英多糖抗幽门螺杆菌相关性胃炎大鼠胃黏膜损伤、保护胃黏膜的作用机制。方法将48只大鼠随机分成正常组、正常+蒲公英多糖组、模型组和模型+蒲公英多糖组,每组12只。模型组和模型+蒲公英多糖组大鼠建立幽门螺杆菌相关性胃炎模型。正常+蒲公英多糖组与模型+蒲公英多糖组给予蒲公英多糖60 mg/(kg·d)灌胃,正常组和模型组用等体积生理盐水灌胃,连续4周。ELISA法检测各组大鼠胃组织中白细胞介素-6(IL-6)、白细胞介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)、前列腺素E 2(PGE 2)含量,Western blot法检测各组大鼠胃组织中诱导型一氧化氮合酶(iNOS)、诱导型环氧合酶(COX-2)、ph-ERK1/2蛋白表达水平。结果模型组大鼠胃组织中IL-6、TNF-α、PGE 2含量及iNOS、COX-2、ph-ERK1/2蛋白表达水平均明显高于正常组及正常+蒲公英多糖组(P均<0.05),IL-10含量明显低于正常组及正常+蒲公英多糖组(P均<0.05);模型+蒲公英多糖组大鼠胃组织中IL-6、TNF-α、PGE 2含量及iNOS、COX-2、ph-ERK1/2蛋白表达水平均明显低于模型组(P均<0.05),IL-10含量明显高于模型组(P<0.05),各指标与正常组及正常+蒲公英多糖组间比较差异均无统计学意义(P均>0.05)。结论蒲公英多糖可通过抑制MAPK/ERK通路减轻幽门螺杆菌相关性胃炎大鼠胃黏膜的炎性反应,从而起到保护胃黏膜的作用。 Objective It is to investigate the mechanism of dandelion polysaccharide(DP)against gastric mucosal injury and protection of gastric mucosa in rats with Helicobacter pylori-associated gastritis in the respect of anti-inflammatory and modulating MAPK/ERK pathway.Methods Forty-eight rats were randomly divided into normal group,normal+DP group,model group and model+DP group,with 12 rats in each group.A model of Helicobacter pylori-associated gastritis was established in the model group and the model+DP group.The normal+DP group and the model+DP group were given DP 60 mg/(kg·d)by gavage,and the normal group and the model group were intragastrically administered with the same volume of normal saline.All the groups were treated for 4 weeks.The levels of interleukin-6(IL-6),interleukin-10(IL-10),tumor necrosis factor-α(TNF-α)and prostaglandin E 2(PGE 2)in gastric tissue of rats in each group were detected by ELISA.The expression levels of inducible nitric oxide synthase(iNOS),inducible cyclooxygenase(COX-2)and ph-ERK1/2 in gastric tissues of rats were detected by Western blot method.Results The contents of IL-6,TNF-α,PGE 2 and the expression levels of iNOS,COX-2 and ph-ERK1/2 in the gastric tissue of the model group were significantly higher than those in the normal group and the normal+DP group(P<0.05),the content of IL-10 was significantly lower than that in the normal group and normal+DP group(P<0.05);The contents of IL-6,TNF-α,PGE 2 and the expression levels of iNOS,COX-2 and ph-ERK1/2 in the gastric tissue of the model+DP group were significantly lower than those in the model group(P<0.05),the content of IL-10 was significantly higher than that in the model group(P<0.05),the differences in these indexes were not significantly different when compared with the normal group and normal+DP group(P>0.05).Conclusion DP can relieve the inflammatory response of gastric mucosa in rats with Helicobacter pylori-associated gastritis by inhibiting MAPK/ERK pathway,thus to protect gastric mucosa.
