摘要
筛选出纯化效果最佳的树脂类型,以提高贝母粗多糖纯化效果。太白贝母和甘孜川贝母粗多糖提取液分别经过DA201-C型、AB-8型、201×4型、HPD600型共4种不同类型树脂静态吸附纯化后,比较其对贝母粗多糖提取液其核酸脱除率、蛋白脱除率,色素脱除率的影响,以及对多糖损失率和羟基自由基清除效果的影响。研究显示201×4型树脂对贝母粗多糖纯化效果最佳,核酸脱除率范围在92.22%~94.41%之间,蛋白脱除率在94.13%~94.95%之间,脱色素率在69.14%~86.07%之间,多糖损失率在62.13%~94.47%之间,羟基自由基清除效果在25.72%~29.82%之间。AB-8型树脂对贝母粗多糖纯化效果次之、其次是DA201-C型和HPD600型大孔树脂。201×4型树脂对贝母粗多糖纯化效果最佳。
In order to enhance the purification efficiency of polysaccharide in Fritillaria (Beimu), the study tried to find out which type of resins was the best one for purification. Four different types (DA201-C , AB-8, 201×4 and HPD600) of resin were studied. The nucleic acid removal rate, the protein removal rate, pigment re-moval rate, polysaccharide loss rate and the hydroxyl free radical scavenging effect were the index. The results showed that the type of 201 ×4 resin was the best one. Its nucleic acid removal rate, the protein removal rate, pigment removal rate, polysaccharide loss rate, the hydroxyl free radical scavenging effect was in the ranges of 92.22%-94.41%,94.13%-94.95%,69.14%-86.07%, 62.13%-94.47%, and 25.72%-29.82%, respec-tively. The purification efficiency of AB-8 resin was the second one, followed by DA201-C and HPD600 resins. The type of 201×4 resin was the best one for purification of polysaccharide in Fritillaria.
