摘要
目的观察还原型谷胱甘肽 (GSH )对帕金森病 (PD)大鼠模型黑质抗氧化水平和线粒体呼吸链功能的影响。方法应用 6 羟基多巴胺 ( 6 OHDA)立体定向注射制作PD大鼠模型。将 2 4只大鼠随机分为 3组 (每组 8只 ) :模型组 ,治疗组 ,假手术组 ,分别给予相应处理 45d ,给药后测定各组大鼠黑质区谷胱甘肽过氧化物酶 (GSH Px)、丙二醛 (MDA)、活性氧 (ROS)及线粒体呼吸链酶复合体Ⅰ水平。结果 ( 1)GSH能使模型大鼠黑质区GSH Px活性增强 ,MDA和活性氧水平降低 ( 11.3 0± 0 .49,1.2 6± 0 .15 ,42 .2 5± 2 .88)。 ( 2 )GSH能增强黑质呼吸链酶复合体I活性 ( 2 90 5 .2 8± 5 2 .15 )。结论GSH能减轻PD模型大鼠黑质区氧化应激损伤 ,并对线粒体呼吸链具有一定保护作用。
ObjectiveTo observe the effects of glutathione on nigral antioxidation system, mitochondrial respiration-chain function of PD model rat.Methods The PD model rats were established through stereotoxic microinjection of 6-OHDA. The 24 rats were randomly divided into 3 groups:Model group(MG), Glutathione group (GG) and sham-operated group (SG). Every group was given relevant treatment for 45 days. The activity of GSH-Px, content of MDA, ROS and mitochondrionase complex I of nigra were assayed after treatment.Results (1)Glutathione could increase the GSH-Px activity and decrease the contents of MDA and ROS of nigral.(2)Glutathione could exert some protective effect on mitochondrial respiration-chain.ConclusionsGSH could decrease the nigral oxidative stress of PD model rats,and exert some protective effect on mitochondrial respiration-chain.
