摘要
目的 研究三氧化二砷 (As2 O3 )对胆管癌细胞的抑制作用及其机制。方法 用不同浓度的As2 O3 处理胆管癌细胞株HCCC - 9810 ,应用倒置显微镜、MTT法、流式细胞仪观察胆管癌细胞的变化。结果 As2 O3 在 0 .75、1.5、3.0、6 .0 μmol/L的浓度对胆管癌细胞作用不同时间 ,作用 72h时生长抑制率分别为 2 2 .34%、4 3.2 5 %、5 9 6 8%、6 9.95 %。结论 适合浓度的As2 O3 在体外可诱导胆管癌细胞凋亡 ,有较明显的时间和剂量依赖性 ,其发生机制可能与细胞周期阻滞有关。
Objective To investigate the apoptosis and mechanism of human biliary tract carcinoma HCCC-9810 cells induced with arsenic trioxide. Methods After treatment with arsenic trioxide, the growth inhibition of HCCC-9810 cells was assessed by MTT colorimetric assay, and their apoptotic rate, by flow cytometry (FCM). Results arsenic trioxide inhibited the growth of HCCC-9810 cells in a certain range of dosage. At the doses of 0.75,1.5,3.0 and 6.0 μmol/L after 72 h of action, the growth inhabitation rate was 22.34%, 43.25%, 59.68% and 69.95%. Conclusion Arsenic trioxide may induce apoptosis of HCCC-9810 cells and there is a liner correalation between apoptosis and the dosage of arsenic trioxide.
