摘要
目的:研究骨髓间充质干细胞定向分化为腱细胞的可能性。方法:通过电穿孔法将外源基因BMP12导入骨髓间充质干细胞中,诱导骨髓间充质干细胞定向分化为腱细胞;在形态学和分子生物学水平上对诱导后的细胞加以鉴定。结果:光镜下,诱导后的细胞形态发生明显改变;RT-PCR结果表明,诱导后的细胞有BMP12和CollagenⅠ的mRNA的表达,而没有CollagenⅢ的mRNA表达;诱导后细胞呈CD44+为98.39%,HLA-DR-。结论:BMP12能够诱导骨髓间充质干细胞定向分化为腱细胞,间充质干细胞有可能成为肌腱组织工程种子细胞来源之一。
Objective: To investigate the feasibility of committed diffe rentiation of bone marrow mesenchymal stem cells (MSCs) into tenocytes induced b y BMP12. Methods: MSCs were transfected with pTARGETTM bearing BMP12 gene by ele ctroporation. The transfected cells were identified by morphological observation and molecular biological measure. Results: Under the observation of light micro scope, the morphology of transfected cells changed significantly comparing to pa rent MSCs. RT-PCR data showed the transfected cells had the expression of BMP12 and CollagenⅠmRNA, but no CollagenⅢ mRNA. The transfected cells were CD44+ 98. 39% and negative for HLA-DR. Conclusion: BMP12 could induce MSCs into tenocytes, and bone marrow MSCs were optional seed cells applying to tendon tissue enginee ring.
