摘要
目的观察生长抑素 (SST)对肝部分切除后裸鼠肝癌组织基质金属蛋白酶 2 (MMP 2 )及其抑制剂 (TIMP 2 )表达的影响。方法 30只裸鼠随机分成 3组 :A组 (对照组 )仅将人肝癌组织种植于右肝叶 ,B组 (肝部分切除组 )和C组 (SST治疗组 ) ,切除肝左叶和中叶后于右肝叶种植人肝癌组织 ,C组术后第 1天开始腹腔注射SST(2 5 0 μg/kg ,2次 /d)B组和A组腹腔注射等量的生理盐水 ,3组动物于 35d后处死 ,检测癌体大小和重量 ,采用免疫组织化学和图像分析法检测肝癌组织微血管密度 (MVD)值、MMP 2和TIMP 2的表达。结果B组癌体重量和体积、MVD和MMP 2表达较A组明显增加 (P <0 0 5 ) ,TIMP 2则明显减低 (P <0 0 5 ) ;C组癌体重量和体积较B组和A组明显减低 (P <0 0 5 ) ,MVD和MMP 2表达较B组和A组明显减低 (P <0 0 5 ) ,TIMP 2表达则较B组和A组明显增加 (P <0 0 5 )结论MMP 2表达上调和TIMP 2表达下调可能为肝部分切除后癌灶新生血管形成增多的原因 ,SST可以通过抑制MMP 2的表达 ,上调TIMP 2的表达 ,从而抑制癌灶新生血管形成而减轻肝部分切除促癌灶生长的作用。
Objective To observe the effect of somatostatin (SST) on the expression of metalloproteinase-2(MMP-2) and tissue inhibitor matrix metalloproteinase-2(TIMP-2) of implanted tumor in nude mice after partial hepatectomy.Method Nude mice were divided into group A(n=10) implanted by human hepatocellular carcinoma(HCC),group B(n=10, partial hepatectomy) and C(n=10,HCC implantation after liver resection). SST(250 μg/kg,b.i.d) was given intraperitoneally in group C. Mice were sacrificed on 35th d, tumor was measured, the expression of MVD.MMP-2 and TIMP-2 was detected by immunochemical staining and quantitative image computer-analysis. Results In group B, the weight and volume and expression of MVD.MMP-2 of tumor tissue was markedly incressed than in group A(P<0.05),the expression of TIMP-2 of tumor tissue was markedly decreased than in group A(P<0.05), in group C, the weight and volume and expression of MVD and MMP-2 of the tumor was markedly decreased than in group B and A(P<0.05),the expression of TIMP-2 was increased markedly than in group B and A (P<0.05). Conclusion SST decreases the expression of MMP-2 and increases the expression of TIMP-2 hence inhibits angiogenesis of tumor tissue.
