脂蛋白脂酶活化剂No-1886对抗脂毒性葡萄糖毒性及对贵州小香猪胰岛细胞的保护作用 被引量:3

摘要 目的 探讨合成化合物No 1 886调节糖、脂代谢的作用机制及对贵州小香猪胰岛结构和功能的影响。方法  3- 4月龄贵州小香猪 1 5只 ,随机选取 5只饲以基础饲料为正常对照组 ;5只饲以高脂高糖饲料为糖尿病对照组 ;5只前 3个月饲以高脂高糖饲料 ,后 5个月饲以高脂高糖饲料加 1 0 %No 1 886为糖尿病治疗组。分别在 0、2、3、4、5、6、8个月末时从禁食过夜的猪眶静脉窦取血样 ,测定血浆葡萄糖、甘油三酯和血清胰岛素浓度。第 8个月末 ,处死动物 ,每只动物取胰腺 1小块 ,1 0 %中性甲醛液固定 ,常规石蜡包埋切片 ,HE染色。另一部分胰腺切片用猪胰岛素抗体作免疫组化染色 ,显微观察照相 ,并用图像分析软件IMAGEJ测量胰岛 β细胞在胰岛中所占的面积。另取 1小块胰腺 ,4℃ ,2 5 %戊二醛固定 ,常规透射电镜样品制备 ,透射电镜下观察并照相。结果 糖尿病对照组和糖尿病治疗组 (加药前 )出现高血糖、高甘油三酯 ,胰岛素水平亦升高 ,正常对照组血糖、甘油三酯、胰岛素未见明显升高。两组比较差异具有显著性意义 (P <0 0 5 ) ;与糖尿病对照组比较 ,糖尿病治疗组加药后血糖、甘油三酯、胰岛素水平降低。从胰腺显微和超微结构观察及 β细胞免疫染色结果看 ,高糖高脂饲料对贵州小香猪胰腺组织结构。 Objective To explore the application mechanism of No 1886, a synthetic compound, adjusting glucose and lipid metabolism and the effect on the structure and function of the islets in diabetic Guizhou minipigs induced by high fat and high sucrose feeding. Methods\ Fifteen Guizhou minipigs about 3-4 months were randomly divided into 3 groups: similar body weight of Normal control diet(CD), high fat high sucrose fed diet(HFSD), and high fat high sucrose fed diet followed by No 1886 treatment (HFSD+No 1886). The fasting concentrations of plasma glucose, triglyceride and serum insulin were analyzed at month 0, 2, 3, 4, 5, 6, and 8 respectively. The animals were sacrificed at the end of month 8 Samples of pancreas were cut and fixed in 10% formalin, paraffin embedded, sectioned and stained with HE. Other sections were stained with an antibody to porcine insulin. After immunostaining, sections were photographed and the percent areas occupied by β cells were measured in all of the three groups by plamimetry with the software IMAGE J. A part of pancreas was fixed with 4℃, 2 5% glutaraldhyde, sampled by means of normal electronic microscope, and observed and photographed under transmit electronic microscope. Results\ High fat high sucrose feeding elevated fasting plasma glucose, triglyceride and serum insulin levels significantly. These values were significantly higher than those in Normal control group ( P <0 05). Supplement of No 1886 into high fat high sucrose diet induced a decrease in plasma glucose, triglyceride and insulin concentration compared with pigs fed with the sole high fat high sucrose diet. The observation of pancreatic islets micro structure and ultra structure and immunostaining results of islet β cells showed that the high fat and high sucrose feeding damaged the pancreatic tissues structure and the structure and function of the β cells. But No 1886 had a protection of pancreatic tissues structure,and the structure and function of β cells. Conclusion\ No 1886 suppressed plasma gluc
