摘要
[目的]研究HBx蛋白对细胞周期的影响及其在肝细胞癌发生中的作用。[方法]采用SP免疫组化方法分别检测HCC组织中HBx蛋白和p21蛋白表达及PCNA。[结果]HCC组织中HBx蛋白的阳性率为52.5%(21/40),阳性信号多呈核型,阳性细胞多呈区域性或簇状分布;p21蛋白阳性表达随HBx蛋白表达的增强而下降;HCC组织中HBx蛋白表达阳性病例PCNA评分高于阴性病例,p21蛋白表达的阳性病例PCNA评分低于阴性病例。[结论]在HCC的发生过程中,HBx蛋白可影响p21蛋白的表达,推进细胞周期,导致细胞增殖和促进肝细胞的恶性转化。
To study the influence of HBx protein to cell cycle and the effect in hepatocellular carcinogenesis.The expressions of HBx protein, p21 proteins and proliferation cell nuclear antigen(PCNA) were detected by SP immunohistochemistry.The positive expression rate of HBx protein was 52.5%(21/40) in hepatocellular carcinoma(HCC).The positive signal of HBx protein was mainly caryotype with regional or clustered cell distribution.The stronger expression of HBx protein, the weaker expression of p21 protein in HCC; The grade of PCNA in HCC with HBx protein positive expression was higher than that with negative expression;but the grade of PCNA in HCC with p21 protein positive expression was lower than that with negative expression.[Conclusions]It suggests that HBx protein may accelerate hepatocyte proliferation and promote malignant transformation of hepatocyte by influencing p21 protein in hepatocellular carcinogenesis.
