摘要
紫芦笋以茎尖为外植体,进行组织培养,其研究结果为:(1)启动培养基:MS+6-BA1.0mg/l;(2)增殖培养基:适合紫芦笋雄株增殖的培养基是:MS+NAA0.5mg/l+6-BA0.5mg/l,每切片平均发芽数可达2.5个;适合紫芦笋雌株增殖的培养基是:MS+6-BA0.3mg/l+NAA0.05mg/l,平均发生芽数可达2.1个;(3)生根培养基:改良MS+IBA1.0mg/l+KT0.2mg/l+NAA0.05mg/l,生根率高达85%;(4)过度移栽技术:选用3条根以上的组培苗在土5份十垤石3份的基质上移栽。
The tissue culture of stem tip from purple Asparagus was studied.The results showed:(1)The suitable medium for induction culture was MS+6-BA 1.0 mg/L;(2)The suitable mutiplication medium for male plant of purple Asparaguswas MS+NAA 0.5 mg/L+6-BA0.5mg/Lwith multiplication coefficient 2.5;The suitable mutiplication medium for famale plant of purple Asparaguswas MS+NAA 0.05 mg/L +6-BA0.3 mg/L with multiplication coefficient 2.1;(3)Rooting medium:Improved MS+NAA0.05 mg/L +IBA 1.0 mg/L+KT 0.2 mg/L with rooting rate 85%;(4)Transition cultivating technique:Tissue culture seedseedings with more than 3 roots cultivated in the mixed soil according to soil/vermiculite rate 5/3.
