摘要
目的:对石菖蒲的几种粗提物的抗抑郁作用进行筛选。方法:选用小鼠尾悬挂试验、大鼠强迫游泳试验和小鼠5-HTP诱导的甩头试验模型对石菖蒲的提取物的抗抑郁作用进行研究。结果:在小鼠尾悬挂试验和大鼠强迫游泳试验中,石菖蒲水提醇沉液与水提液一样均能使动物的不动时间显著缩短,两者作用与抗抑郁药氟西汀相当。石菖蒲的乙醇提取液也与水提液一样能显著缩短小鼠的不动时间,与氟西汀阳性对照组比较,石菖蒲乙醇提取液组的作用与氟西汀无显著性差别,水提液组作用稍差。石菖蒲醇提后的其他萃取物(正丁醇、氯仿、乙酸乙酯)在行为绝望动物模型上的作用较小或无效。在小鼠5-HTP增强甩头试验中,较大剂量(10g/kg)的石菖蒲水提醇沉液与氟西汀一样能增加小鼠甩头反应次数。结论:石菖蒲抗抑郁成分的有效部位主要存在于水提取液和乙醇提取液中,其抗抑郁作用可能与阻断中枢5-HT等单胺类递质的重摄取有关。
Objectice: To assess the antidepressant effect of the several crude extracts of Rhizoma Acori Tatarinowii . Methods: The antidepressant effect of the several crude extracts of Rhizoma Acori Tatarinowii was evaluated with rat force swimming test, mice tail suspension test and 5-HTP induced head-twitches model in mice. Results; Similar to fluoxetine, the aqueous extract and ethanolic precipitate of aqueous extract of Rhizoma Acori Tatarinowii significantly decreased the accumulative immobility time in rat forced swimming test and mice tail suspension test. The ethanolic extract and the aqueous extract of Rhizoma Acori Tatarinowii have similar effect in shortened immobility time in mice tail suspension test. The other ( n-butanol, chloroform, acetic ester) extracts of Rhizoma Acori Tatarinowii produced a little or less antidepressant effect on the behavioural despair animal model of depression. Similar to fluoxetine, high dose (10 g/kg) of the ethanolic precipitate of aqueous extract of Rhizoma Acori Tatarinowii significantly increased the times of 5-HTP induced head - twitches in mice. Conclusion: The active ingredients with antidepressant action of Rhizoma Acori Tatarinowii mainly exist in the aqueous extract and ethanolic extract of Rhizoma Acori graminei . The mechanism of antidepressant action of the extracts of Rhizoma Acori Tatarinowii may be related to the blocking of serotonin re-uptake in central nervous system.
