摘要
目的:研究柴胡(Bupleurum chinense DC.,BCDC)醇提液、正丁醇提取部位(DC-Bu)和水提取部位(DC-Aq)对抑郁模型大鼠脂质过氧化及免疫功能的影响。方法:将42只雄性SD大鼠随机分为7组,分别为正常组、模型组、DC-Bu高低剂量组(200、100mg/kg)、DC-Aq高低剂量组(200、100mg/kg)及氟西汀组(20mg/kg)。除正常组,均接受21d慢性应激,造成大鼠抑郁模型,每次应激前1h灌胃给药,正常组及模型组给予等量的含0.01%吐温80的蒸馏水。给药22d后,测定大鼠血清谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量。给予正常小鼠DC-Bu或DC-Aq,采用MTT法检测含药血清对小鼠脾淋巴细胞增殖的影响。结果:DC-Bu及DC-Aq各剂量组均可提高慢性应激抑郁大鼠血清GSH-Px、CAT、T-SOD活力,降低慢性应激抑郁大鼠血清MDA含量,含20%DC-Bu血清和含10%,20%DC-Aq血清对小鼠脾淋巴细胞有增殖作用。结论:DC-Bu和DC-Aq的抗抑郁作用可能与增强慢性应激抑郁大鼠的抗氧化功能有关;其含药血清具有增强免疫功能的作用。
Aim:To investigate the effects of the butanol and aqueous fractions obtained from the 60% ethanol extract of Bupleurum chinense DC. (BCDC) on lipid peroxidation in chronic unpredictable mild stress (CUMS) model of depression in rats, and lymphocyte proliferation in mice. Methods:42 SD rats were divided into 7 groups, the control group, the model group, DC-Bu groups(200, 100 mg/kg), DC-Aq groups(200, 100 mg/kg)and fluoxetine group(20 mg/kg). The model was reproduced by chronic unpredictable mild stress (CUMS) and separation for 21 d. Rats received perfusion lh before model establishing, the control group and the model group were perfused with 0.01% tween 80 solution. The serum was separated at the 22 d to detect the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) and the content of malondialdehyde (MDA) in rat serum. The effect of the serum of mice treated with aqueousand fraction or butanol fraction on the lymphocyte proliferation in vitro were determined by MTT assay. Results: DC-Bu and DC-Aq could obvionsly enhance the activities of SOP GSH-Px and CAT at dose 200mg/kg,DC-Bu(200 mg/kg) and DC-Aq(200 mg/kg, 100 mg/kg) significandy reduced the MDA content in serum in CUMS rats; DC-Bu-S and DC-Aq-S concentration-dependently increased ConA-induced proliferation of spleen lymphocytes, and 20% concentration of serum had better effect with significant differences ( P 〈 0.05 or P 〈 0.01). Conclusion: The possible mechanism of DC-Bu and DC-Aq for anti-depressive action may be involved in anti-oxidation; the drug serum could reinforce immune function,combination with antidepressant may improve the antidepression effect.
