摘要
〔目的〕本研究针对转基因微生物扩散与外源基因转移的途径和方式 ,建立转基因微生物扩散与外源基因转移试验模型 ,并对该模型的灵敏度和准确性等技术指标进行了验证。〔方法〕应用该试验模型 ,监测转抗菌肽CAD基因酵母饲料添加剂在饲喂鸡、虾和猪过程中的扩散与转移。〔结果〕结果表明 ,试验模型能有效地监测抗菌肽CAD基因酵母活菌的扩散 ,及其抗菌肽CAD基因、nptⅡ抗性标记基因和亮氨酸补偿型报告基因的转移。〔结论〕该模型可用于监测转基因微生物向环境的扩散及其外源基因向受试动物胃肠道微生物、环境微生物及受试动物组织的转移。
Diffusion of GM microorganism to the environment and transferring of its foreign gene to non-target organisms during application of GM microorganism are important items for evaluation of safety of the GM microorganism and its products.Based on the approach and mode of diffusion and transfer,we established a model of GM microorganism diffusion and its foreign gene transferring and evaluated its sensitivity and accuracy.Using this model,the diffusion and transfer of GM yeast feeding additive with Cecropin AD gene had been inspected when it was used to feed chickens,shrimps and pigs.The results showed that the model can be used to effectively detect the diffusion of the GM yeast and transfer of Cecropin AD gene,npt Ⅱ marker gene and Leucine-compensating report gene.The model is available to detect the spread of GM microbe in the environment and the transfer of its extrinsic gene to microorganisms in the environment as well as microbes in the animal gastrointestinal tract and animal tissues.
