摘要
用0.15μg/mlPHA 诱导 T 细胞活化作模型,发现 TⅡ(5μg/ml)和 Bt_2cAMP(500μm)均能抑制 T 细胞从 Go 相进入 G_1a 相和从 G_1a 进入 G_1b 相,并抑制活化抗原 Tac、Ia、T(?) 和4F2的表达.TⅡ和Bt_2cAMP 还抑制活化 B 细胞在外源性 IL—2诱导下进入 S 相和 G_2/M 相.另外,T Ⅱ和 Bt_2cAMP 还抑制 T细胞在1.5μg/mlPHA 诱导下产生 IL-2。TⅡ和 Bt_2cAMP 的上述作用十分相似.有趣的是,TⅡ和 T细胞一起培养30小时,T 细胞中 cAMP 量明显增多。cAMP 增加程度与 TⅡ剂量正相关。上述结果提示,TⅡ对 T细胞增殖的抑制作用至少部分经由 cAMP.
It has been observed in human T cells activted with 0.15μg/ml of PHA that both T Ⅱ(5μg/ml)and Bt_2cAMP(500μM)inhibites the entrance of T cells from GO into G_(1a)phase and from G_(1a)into G_(1b)phase,and inhibites the expression of activation antigens Tac,Ia T_9 and 4F2.Both T Ⅱ and BT_2cAMP also suppress the entrance of activated T cells into S and G_2/M phase in response to exogenous IL—2,and suppress IL—2 production of T cells stimulatde with 1.5μg/ml of PHA.The suppressive effect of T Ⅱ and Bt_2cAMP is quite similar.Interestingly,T■can induce cAMP increase in T cells in a dose—dependent manner.The data suggests that the suppressive effect of T■on T cell proliferation might be(at least partially)mediatde by cAMP.
