低浓度肝素对重组人血小板型磷脂酶A_2杀菌活性的抑制 被引量:2

摘要 目的 :研究肝素对重组人血小板型磷脂酶 A2 (PL A2 )杀菌活性的影响 ,探讨血小板型 PL A2 电荷性质在其杀菌机制中的作用。方法 :将金黄色葡萄球菌与重组人血小板型 PL A2 共同孵育 ,试验组加入肝素 ,通过测定细菌菌落数来评价杀菌效果。结果 :低浓度肝素对重组人血小板型 PL A2 杀菌功能有明显抑制作用 ,仅 0 .0 30 0 U / ml的肝素其杀菌抑制率达 10 0 %。结论 :1血小板型 PL A2 的正电荷性在其杀菌作用中是关键因素 ;2肝素是血小板型 PL A2 一种有效的抑制剂。 Objective:To study the effects of heparin on the bactericidal activity of recombinant human platelet phospholipase A 2(PLA 2) , and to explore the role of charge properties of platelet PLA 2 in the bactericidal mechanism of the PLA 2.Methods:Staphylococcus aureus was incubated with recombinant human platelet PLA 2,while the experiment group was added with heparin. Antibacterial activity was determined by counting colony form units of bacteria.Result:Heparin in low concentration significantly inhibitted bactericidal activity of recombinant human platelet PLA 2, the inhibition rate of 0.03U/ml heparin in 100%.Conclusion:①Cationic property of the enzyme is key factor in bactericidal activity of the PLA 2;②Heparin is an effective inhibitor of platelet PLA 2.
