摘要
目的:改进分离培养肌肉卫星细胞的方法。方法:选择兔前肢三头肌通过胶原酶消化、提纯肌束,再用胰蛋白酶消化肌束上的卫星细胞。在含有200ml/l胎牛血清的培养液中培养,使用骨骼肌肌动蛋白抗体进行鉴定。结果:使用本技术可以得到高纯度的肌肉卫星细胞(>0.99),骨骼肌肌动蛋白和肌球蛋白可以用于卫星细胞的鉴定。结论:使用分离纯化肌束的方法,有效的避免了成纤维细胞的污染,从而得到高纯度卫星细胞,方法简单,可以用于今后的研究工作。
Objective To improve the method of isolation the muscle satellite cells. Methods The muscle fibers abstained from rabbit. With the modified method of Rosenblatt , the muscle sample was digested with collagenase-I first, in order to get purified muscle fibers. The secondly, muscle fibers were digested with trypsin to get satellite cells. Skeletal muscle satellite cells identified with cellular immunochemical stain. Results Using this method, the purification of satellite cells is over 0.99, satellite cells were positive and myotubes were strong positive by cellular immunochem stain with á-sarcometricaction. Conclusion The modified method can produce high purification satellite cells with simple process.
