摘要 白腐担子菌Trametes versicolor在加入0.05%Tween 80,(NH4)2-tatrate和80 mg/kg Mn-SO4的Kirks培养基上能够高活性地分泌Mn-过氧化物酶。在该培养基中培养10 d,活性达到最高。通过DEAE-Sepharose CL-6B,SP-Sepharose和离子交换柱分离纯化,获得2种类型的Mn-过氧化物酶同工酶,并对这2种酶的酶动力学,N-端的氨基酸序列及其编码基因的核酸序列做了研究。 The white-rot basidiomycete Trametes versicolor produced high levels of Mn-peroxidase activity when grown on Kirks media with 0. 05% Tween 80, 2. 4 mmol/L (NH4)2-tatrate, 80 mg/kg of MnSO4. Maximum activity of Mn-peroxidase was appeared on the 10th day of culture on above medium of combinations. Two types of isoenzymes of Mn-peroxidase were separated by DEAE-Sepharose CL-6B, SP-Sepharose, anion and cation exchange chromatography on Mono-Q and Mono-S column. Those iso-enzymes were characterized in view point of enzyme kinetics, N-terminal amino acid sequences, and base sequences of the coded genes, etc.
