酒精性脂肪肝大鼠模型的建立及新清肝饮煎剂的防治作用 被引量:12

摘要 目的 :探讨建立酒精性脂肪肝 (alcoholicfattyliver,AFL)动物模型的方法及中药新清肝饮煎剂的防治作用。方法 :大鼠用白酒和玉米油混合物灌胃 1次 /d ,连续 6周 ,观察其肝脏病理改变及血脂变化 ,并观察中药新清肝饮煎剂的防治作用。结果 :模型组大鼠肝脏病理表现为广泛的脂肪变性 ,血清甘油三酯 (triglyceride ,TG)、总胆固醇 (totalcholesterol ,TC)升高 ,与正常组比较有显著差异 (P <0 .0 1) ;丙氨酸氨基转移酶 (alanineaminotransferase ,ALT)、天冬氨酸氨基转移酶 (aspartateaminotransferase ,AST)升高 ,与正常组比较有显著差异 (P <0 .0 5)。新清肝饮组肝脏脂肪变性明显减轻 ,血清TG、TC、ALT、AST与模型组比较明显下降 (P <0 .0 1)。结论 :白酒加玉米油灌胃建立大鼠AFL模型 ,与AFL的发病相似 ,便于药物干预实验。 Objective:To establish an animal model of alcoholic fatty liver (AFL) and to observe the p revention and treatment with New Qinggan Decoction (NQGD). Methods: Female SD rats were exposed to the mixture of white spirit and corn oil contin uously for up to 6 weeks via an intragastric feeding protocol. The pathologic al teration of the liver and the changes of serum lipid were observed, and the effe ct of NQGD for preventing and treating AFL was studied. Results: W ide hepatic steatosis was induced in model group rats. Serum triglyceride (TG) a nd total cholesterol (TC) of the model group were higher than those of the norma l group (P<0.01). Serum aspartate aminotransferase (AST) and alanine aminotr ansferase (ALT) of the model group were higher than those of the normal group ( P<0.05). In NQGD group, hepatic steatosis was milder than that in the model group, and serum TG, TC, AST, ALT descended significantly as compared with the m odel group (P<0.01). Conclusion: AFL model of rats established by feeding the mixture of white spirit and corn oil is similar to the episode o f the human AFL, and is easy to intervene with medicine. NQGD can prevent the oc currence of AFL in this model.
