摘要
目的:探讨不同时段缺血对血管组织超微结构的影响。方法:以15%DMSO为低温保护剂,采用两步冷冻法,对SD大鼠血管进行冷藏缺血和常温缺血实验,时间为1、2、3、4、5、6和8h。动脉标本冷冻维持温度为-55℃,静脉为-60℃,-196℃液氮保存1W。复温后标本透射电镜观察。结果:缺血8h,冷藏组血管组织的内皮细胞轻度肿胀,胞浆内线粒体嵴稀疏或破坏。常温组血管组织的内皮细胞坏死增多,胞膜、核膜破裂,细胞核破坏,残留胞膜,内弹性膜区域性裸露等。结论:冷藏缺血,血管组织的超微结构损伤轻微,表明冷藏能明显延长血管组织的缺血耐受时间。
Objective: To investigate the effect of different ischemia periods on the ultrastructure of SD rats. Methods: With 15% DMSO as low temperature protective reagent, cold storage ischemia and normal temperature ischemia experimens on SD rat vessel tissues were conducted, the duration of which was 1h,2h,3h,4h,5h,6h and 8h.The artery tissue was given - 55℃ holding temperature and the vein tissue - 601 holding temperature. The cold pretreatment in liquid nitrogen lasted for one week.The tissue was sliced and studied by TEM( transmission electrical microscope). Results: The vessel tissue endothelium cell of the cold storage group of 8 - hour ischemia was found slight swelling and the mitochondrial crista of cytoplasm arefaction or destruction. The vessel tissue endothelium cell of the normal temperature group was associated with more and more nerosis broken cell membrane and nuclear membrane, destroyed nucleus remaining cell membranes and exposed regional internal elastic membranes. Conclusion: The damage caused by cold storage ischemia to the ultrastructure of vessel tissue is slight and the cold storage can prolong the duration of tissue endurance for ischemia.
