摘要
目的观察普鲁托品(Protopine,Pro)对气管平滑肌的影响。方法豚鼠离体气管条法和整体药物引喘法。结果 ①Pro能明显抑制组织胺、乙酰胆碱引起的豚鼠离体气管收缩,最大反应压低呈非竞争性拈抗,与氨茶碱(Ami)作用相似。pD2’分别为3.76和4.16,Pro与Ami合并应对组胺引起豚鼠离体气管条收缩具有协同的抑制作用。②Pro 16mg/kg、32mg/kg、64mg/kg腹腔注射剂量依赖性延长组胺诱发的引喘潜伏期,抽搐动物数亦随剂量增加而减少,与氨茶碱作用相似,但二药合用与一药单用相比.其引喘潜伏期无显著性差异。结论普鲁托品可抑制豚鼠气管平滑肌的收缩并能产生一定的平喘效果。但与氨茶碱合用,其协同作用尚不能肯定。
Aim The effects of protopine(Pro) on tracheal smooth muscle were observed. Methods The methods of isolated guinea pig trachea and the animal's model asthma in conscious guinea pigs were used. Results (1) Pro resembles as aminophyiline(Ami), it markedly depress the tracheal spiral strips constriction of guinea pig induced by histamine or acetylcholine, The maxmal response was signigtcantly depressed, showing a non-competitive antigonism.The pD2'of Pro for constrictions induced by Histamine and Acetylcholine were 3.76 and 4.16, respectively. Pro in combination with Ami can produce synergic depressive action for constrictions of tracheal spiral strips of guinea pig induced by histamine. (2)In the asthmatic model of guinea pig induced by inhalation of 0. 1 % histamine phosphate aerosol mist for ten seconds, Pro given peritoneally in dose of 16mg/kg, 32mg/kg and 64mg/kg, prolonged the latent period of asthmatic attack and decreased the numbers of asthmatic attack animals in dose-dependently. But no difference significantly could be seen in latent period for asthmatic attack between Pro used alone and combined with Ami. Conclusions Pro can inhibit the constriction of tracheal smooth muscle to induce an antiasthmatic effect, but the synergistic effect between Pro and Ami is not sure.
