摘要
本文报道了胚胎大鼠脊髓腹侧神经细胞体外培养方法,应用相差显微镜、电子显微镜以及细胞化学染色等观察神经细胞的生长分化,并以神经细胞数、神经突起数及其长度为指标比较某些神经营养因子对神经细胞生长分化的作用.结果表明,培养的神经细胞中多数为运动神经元.大鼠脊髓的条件培养液、小鼠骨胳肌的条件培养液、孕酮和胰岛素等对培养的胎鼠脊髓腹侧神经细胞的生长分化具有促进作用.
A method of culture of nerve cells isolated from fetal rat ventral spinal cord(VSC) was described.The maturation and differentiation of these cells were studied with phase-contrast microscope,electron microscope and cytochemical staining techniques.Methods for estimating nerve cells,nerve cell process and its length were established.By comprehensive analysis of the results,we found that most multipolar nerve cells from VSC were motoneurons.Furthermore we studied the effect of some neurotrophic factors on the differentiation and maturation of VSC cells in culture.The results showed that rat spinal cord conditioned medium,mouse skeletal muscle conditoned medium,progesterone and insulin all promoted the maturation of VSC nerve cells in culture.
