基于网络药理学研究人参、川芎药对有效成分抗动脉粥样硬化的作用机制 被引量:19

Mechanism of Effective Constituent Renshen(Radix Ginseng) and Chuanxiong(Rhizoma Ligustici Chuanxiong) Drug Pair in Anti-atherosclerosis: Based on Network Pharmacology

摘要 目的探讨人参、川芎药对有效成分抗动脉粥样硬化的作用机制。方法通过PubMed、CNKI、中药系统药理学数据库和分析平台TCMSP数据库检索人参总皂苷、川芎嗪化学成分,构建化学成分数据库。通过BATMAN-TCM平台预测成分靶标,并预测疾病靶标,利用Cytoscape 3. 6. 1软件进行核心靶标筛选、GO基因及KEGG通路富集分析,构建药物成分-作用靶标-代谢通路网络图,探讨其抗动脉粥样硬化的作用机制。结果本研究收集化学成分17个,筛选成分-靶标PPI网络有464个节点,214832条连接。药物-疾病核心靶标17个,GO基因富集分析提示涉及生物过程、分子功能2个方面,KEGG通路富集分析潜在调控通路有PPAR通路、脂肪细胞因子信号通路及胆汁酸分泌,且发掘靶标、通路中多个与肥胖相交叉,其中PPARγ/PGC-1α/UCP1信号通路能够通过促进白色脂肪棕色化发挥抗动脉粥样硬化作用。结论人参、川芎有效成分可能通过PPARγ/PGC-1α/UCP1信号通路促进白色脂肪棕色化,改善动脉粥样硬化。 Objective To investigate action mechanism of effective constituent Renshen(Radix Ginseng)and Chuanxiong(Rhizoma Ligustici Chuanxiong)drug pair in anti-atherosclerosis.Methods The chemical constituents of total saponins of panax ginseng and ligustrazine were searched in PubMed,Chinese National Knowledge Infrastructure(CNKI),traditional Chinese medicine system pharmacology database and analysis platform(TCMSP database)to construct a chemical composition database.The component targets and disease targets were predicted through the BATMAN-TCM platform.Cytoscape 3.6.1 software was used for core target screening,GO gene and KEGG pathway enrichment analysis,and a network map of drug component target metabolic pathways was constructed to explore its action mechanism of anti-atherosclerosis.Results In this study,17 chemical components were collected,and the target component of the screening PPI network had 464 nodes and 214 832 connections.There are 17 core targets for drug diseases.The analysis of GO gene enrichment suggested two aspects of biological processes and molecular functions.The KEGG pathway enrichment analysis has potential regulatory pathways including PPAR pathway,adipocytokine signaling pathway and bile acid secretion,and explored that multiple of targets and pathways were intersect with obesity,and the PPARγ/PGC-1α/UCP1 signaling pathway can exert an anti-atherosclerotic effect by promoting browning of white fat.Conclusion The effective constituent Renshen(Radix Ginseng)and Chuanxiong(Rhizoma Ligustici Chuanxiong)can improve atherosclerosis through PPARγ/PGC-1α/UCP1 signaling pathway promoting browning of white fat.
